Pleiotropic Roles of Calmodulin in the Regulation of KRas and Rac1 GTPases: Functional Diversity in Health and Disease

Abstract: Calmodulin is a ubiquitous signalling protein that controls many biological processes due to its capacity to interact and/or regulate a large number of cellular proteins and pathways, mostly in a Ca2+-dependent manner. This complex interactome of calmodulin can have pleiotropic molecular consequences, which over the years has made it often difficult to clearly define the contribution of calmodulin in the signal output of specific pathways and overall biological response. Most relevant for this review, the abil… Show more

“…Phagocytosis is a dynamic, multi-step process regulated by signaling. Meth significantly increased calcium signaling effectors, calmodulin ( CALM1/2/3 ) and CAM2KD , which activate the proteins that contribute to cell motility and contraction, such as PRKCA ( Figure 2 D) [ 54 , 55 ]. Membrane lipids enriched for PI(3)P anchor proteins that promote trafficking of phagocytic vesicles [ 50 ].…”

“…(28) CaM is the most widely distributed, most functional, and most studied calcium signaling receptor in cells and is closely related to many cell functions such as regulating cell division, cyclic nucleotide metabolism, and DNA synthesis. (29) CaM is a small soluble globulin molecule consisting of 19 amino acids and is known to be resistant to heat and acid. (30) Due to its strong affinity for Ca 2+ , CaM is the most important calcium receptor in cells.…”

“…The reciprocal expression of these two proteins is further supported by studies showing that RasGRF2 is activated by a surge in intracellular Ca 2+ [ 20 , 100 ] and that this is accompanied by its degradation in proteasomes [ 101 ]. The activity of RasGRF2 is closely linked to Ca 2+ homeostasis, in that elevated Ca 2+ levels stimulate calmodulin-mediated RasGRF2 activation via its IQ domain, with subsequent activation of Ras and/or Rac1 mediated MAP kinase signaling [ 102 ]. Overexpression of AnxA6 in TNBC cells not only blocked the EGF/EGFR or Ca 2+ ionophore stimulated a surge in intracellular Ca 2+ levels, but also the degradation of RasGRF2 [ 20 , 100 ].…”
