Platinum Sensitivity–Related Germline Polymorphism Discovered via a Cell-Based Approach and Analysis of Its Association with Outcome in Ovarian Cancer Patients

Abstract: Purpose Utilizing cell-based approaches to identify genetic markers predictive of patients’ risk for poor response prior to chemotherapy. Experimental Design We performed genome-wide association studies (GWASs) to identify SNPs associated with cellular sensitivity to carboplatin through their effects on mRNA expression using International HapMap lymphoblastoid cell lines (LCLs) and replicated them in additional LCLs. SNPs passing both stages of the cell-based study were tested for association with progressio… Show more

“…Another taxane, paclitaxel, reportedly induces peripheral neuropathy, and several laboratories have investigated the association between SNPs and neurotoxicities [26,27]. Our present results may be useful for choosing safer taxanes, docetaxel or paclitaxel, for patients who are treated for gynecological cancers as well as other types of malignancy.…”

“…Apoptosis can be induced in LCLs by pharmacologic agents, such as the chemotherapeutic drug carboplatin (20,32). Apoptosis induced by carboplatin is at least partially caspase-9-dependent (33), although caspase-8 may also be involved (34).…”

“…Pinpointing a subset of very poor prognosis patients prompts consideration of additional treatment options that may improve survival for these patients. Studies of recurrence and platinum response are affected by the different definitions of tumor recurrence being used at different institutes (35). It is possible that greater prediction accuracy would be achieved when a standardized definition of progression is applied to all cases in the validation set.…”
