Platinum concentration in human tumors of head and neck, uterine cervix, and breast following treatment with cisplatin

Abstract: Intratumoral platinum concentrations were measured in three tumor sites (head and neck, uterine cervix, and breast) 48 h after cisplatin administration according to the same protocol. The platinum levels were in the same order of magnitude in all tumors, but the concentration in breast tumors was found to be higher than that in tumors of the head and neck and of the uterine cervix.

“…Platinum concentrations have been measured in biopsies of tumours of the head and neck after intravenous and intra-arterial infusions of cisplatin. The platinum concentrations found were in the range 0.71 to 7.9 ,ug g tumour (Gouyette et al, 1986;Hecquet et al, 1985).…”

“…In order to quantify the activity content in the tumours, liver and kidneys in the patients, phantom studies were performed. A known activity of 191 Pt was filled in spheres with diameters of 15,20,25 and 30 mm simulating tumours, a MIRD (16) liver phantom and a kidney phantom consisting of two cylinders, 50 mm in diameter and 62 mm in length. The 'organs' were inserted in a water-filled, elliptical thorax phantom (adult MIRD (16)).…”

“…Cisplatin (cis-diamminedichloroplatinum) was developed by Rosenberg (36) in the 1960s and was initially used in the treatment of head and neck, uterine and bladder cancers (37). Cisplatin is one of the most important drugs for the treatment of gastric cancer; it has demonstrated a high positive-response rate in the treatment of gastric cancer.…”
