Platelet transmembrane signalling responses to collagen in familial hypercholesterolaemia

Abstract: Abstract.Washed platelets from patients with familial hypercholesterolaemia (FH) were found to be more reactive towards collagen than those from control subjects. The dose required to achieve half maximum aggregation was found to be 0.6 ml-' for FH patients whilst that for control subjects was 1.25 pg ml-'. In both types of platelet, intracellular Ca2+ levels, as monitored by the Ca2+-dependent photoprotein, aequorin, rose on stimulation with collagen and then fell to basal levels, probably due to resequestrat… Show more

“…Briefly, platelet rich plasma was adjusted to a pH of 6·5 – to prevent fibrinogen binding to its receptor on platelets GPIIbIIIa – and co‐incubated with two units of apyrase per millilitre to prevent low‐level ADP‐stimulation during preparation. Platelets were pelleted and washed twice with an EGTA‐containing HEPES‐buffer (Cooper et al , 1994) supplemented with apyrase. Platelets were finally resuspended in HEPES‐buffer lacking EGTA and apyrase.…”

“…The involvement of platelet-LDL interactions herein is illustrated by observations in FH patients whose platelets are hyperreactive. This is demonstrated by increased ␣ -granule secretion [2,7] , increased superoxide anion production [8] , increased fi brinogen binding [9] and platelet aggregation [2,7,10] after stimulation. In addition, platelets have been found in the circulation of FH patients [11,12] and animals [13] in an activated state.…”

“…Blood was anticoagulated with ACD and processed within 1 hour of collection. Washed platelets were separated from platelet rich plasma as previously described [25] and platelet count was adjusted to 200×10 9 l -1 by suspending in HEPES buffered saline (HBS), containing NaCl (140 mM), KCl (2.7 mM), Glucose (5mM), BSA (1mg/ml), CaCl 2 (1mM), MgCl 2 (1 mM) and HEPES (10 mM) (pH 7.3).…”
