Platelet-targeted fibrinolysis enhances clot lysis and inhibits platelet aggregation.

Abstract: Therefore, the targeting of urokinase to the GP IIb/IIIa platelet receptor both accelerates clot lysis (when platelets are associated with a fibrin clot) and inhibits platelet aggregation.

“…The SK activity was determined according to the previously reported method (Bode et al, 1991;Couto et al, 2004) with a slight modification. Entrapment efficiency was calculated by the following equation: C/T Â 100 (C: amount of SK incorporated into liposome, T: total amount of SK).…”

“…Thrombin is preferentially expressed on the shell of nascent thrombi, where platelet turnover is occurring, whereas in preexisting clots, any residual thrombin is sequestered within the core, where it is less accessible to platelets (18). Drugs used in previous attempts to target uPA to platelets have lacked the thrombin activation mechanism, had short circulating half-lives, and failed to avoid systemic fibrinolysis (19).…”

“…32 Despite increasing the potency by fusing PAs to monoclonal antibodies or Fab fragments, chemical conjugation has several disadvantages, including the inactivation of enzymatic sites by the harsh chemical conditions for conjugating, which can reduce efficacy. 31,33 In addition, the large size of the antibody fusion constructs hampered tissue penetration.…”
