Platelet Nitric Oxide Metabolites in Migraine

Abstract: Nitric oxide (NO) is a candidate as a causative molecule in migraine. We determined nitrite, total nitrate/nitrite, and cyclic guanosine 3',5'-monophosphate (cGMP) concentrations in platelets from 30 migraine without aura (MwoA) patients and 17 migraine with aura (MwA) patients. All migraine patients were studied during their migraine attacks. The control group consisted of 28 healthy volunteers. Concentrations of platelet nitrite and total nitrate/nitrite were determined using simple and sensitive nitrate/nit… Show more

“…Therefore, we measured total nitrite and nitrate levels as an indicator of NO production. Similar to previous studies (D 'andrea et al 1994;Sarchielli et al 1996;Shimomura et al 1999), we found that platelet nitrite and nitrate levels were significantly higher in migraine patients during their migraine attack. Shimomura at al.…”

“…Error bars show S.D. ), the best index of overall NO production is the total concentration of both nitrate and nitrite (Shimomura et al 1999). Therefore, we measured total nitrite and nitrate levels as an indicator of NO production.…”

“…Additionally, it is suggested that intranasal hydroxocobalamin, a NO scavenger, may have a prophylactic effect in migraine [22]. Highly increased NO levels of our migraine patients during attack and interictal period as compared to the controls are in concordance with the results of the most studies [16][17][18][19][20].…”

“…This increase may account for some alterations in platelet function observed in migraineurs, such as the increase in platelet content of L-arginine and the reduced responsiveness to collagen; the latter finding was first shown by D'Andrea et al [5] and later confirmed by our group [4,30]. A recent study in migraine patients confirmed the activation of NOS in platelets, especially during attacks, by demonstrating increased NO end-products compared with headache-free period [31].…”
