Platelet IgG Fc receptor

Abstract: The glycoprotein localization of the platelet binding site for the Fc IgG has been the subject of debate. We attempted to resolve this issue by relating the binding of radiolabeled IgG immune complexes composed of heat-aggregated IgG to platelets from healthy individuals; an individual with Bernard-Soulier syndrome lacking glycoproteins IIb and IX; and a patient with Glanzmann's thrombasthenia lacking glycoproteins IIb and IIIa. The binding of IgG complexes to platelets was determined by measuring the specific… Show more

“…It is generally accepted that ITP reflects accelerated destruction of platelets sensitized by autoantibodies or antibody-containing immune complexes (Kelton et al, 1987;Rosenfeld et al, 1985). Autoantibodies to platelet glycoproteins (GP) IIb/IIIa and Ib/IX have been reported in ITP (Beardsley et al, 1984;Woods et al, 1984a, b;Berchtold & Wenger, 1993;Berchtold et al, 1989).…”

“…Platelets express Fc␥RIIA, a receptor for IgG, which mediates the recognition of immune complexes and IgG-coated targets (18,21,22,31). In contrast to leukocytes that express a variety of Fc␥Rs, platelets express only Fc␥RIIA.…”

“…Radioimmunoprecipitation studies. This technique was performed as described (Kelton et al, 1987) except that the platelets were labelled with 12SIodine using lactoperoxidase. Labelled platelets (at a concentration of 2 x 108/ml) were solubilized using 1% Triton X-100 in 20 mmol/l Tris, 100 mmol/l NaCl pH 7.4 containing 0.1 mmol/l leupeptin (Sigma Chemical Corp., St Louis, Mo.…”

After centrifugation to clear insoluble material, 1 ml of lysate was used for immunoprecipitation as described (Kelton et al, 1987). 4. lmmunodepletion studies.…”
