Platelet-derived Growth Factor Receptor-induced Feed-forward Inhibition of Excitatory Transmission between Hippocampal Pyramidal Neurons

Abstract: Growth factor receptors provide a major mechanism for the activation of the nonreceptor tyrosine kinase c-Src, and this kinase in turn up-regulates the activity of N-methyl-D-aspartate (NMDA) receptors in CA1 hippocampal neurons (1). Unexpectedly, applications of platelet-derived growth factor (PDGF)-BB to cultured and isolated CA1 hippocampal neurons depressed NMDA-evoked currents. The PDGF-induced depression was blocked by a PDGF-selective tyrosine kinase inhibitor, by a selective inhibitor of phospholipase … Show more

“…Although NMDA receptors themselves are known to be both positively and negatively regulated by signaling pathways downstream of PLC, such as PKC, inhibition of PLC with U73122 on its own has no effect on NMDA receptor currents (Lei et al, 1999;Holohean and Hackman, 2004). Thus, U73122 inhibition of PLC is more likely preventing LTD induction by blocking signaling pathways downstream of NMDA receptors.…”

“…In contrast, LTD of the NMDAR component at CA1 synapses is independent of clathrin and instead requires an intact actin cytoskeleton. The inhibition of NMDA-mediated currents in CA1 pyramidal neurons by PDGF-BB is also very long lasting and depends upon an intact actin cytoskeleton (17,18), however, it is not clathrin-dependent but is also blocked by inhibitors of serine-threonine phosphatases (17). Therefore it closely resembles LTD of NMDA synaptic currents.…”

“…Application of PDGF-BB to isolated or cultured hippocampal neurons caused a long-lasting decrease in peak NMDA-evoked currents (17,18). To test if the inhibition of NMDA receptor currents involved NR2B over NR2A receptors we applied NR2 subunit-selective antagonists to the bath (27,28).…”

“…Similarly, PDGF-A and -B and their receptors (PDGFRa and PDGFRb) have a variety of roles in the brain. These include proliferation, migration, and differentiation of oligodendrocytes, neurite outgrowth (45), modulations of NMDA-receptor function (46), and neuroprotection (47). Elevation of these factors improves cognitive deficits and mental activity in aged animals (48).…”
