Platelet-derived growth factor promotes proliferation of erythropoietic progenitor cells in vitro.

Abstract: A B S T R A C T To investigate serum requirements for optimal erythropoiesis in vitro, we studied the response of erythroid progenitor cell proliferation in culture to platelet-derived growth factor (PDGF). Human bone marrow cells cultured with platelet-poor plasma-derived serum (PDS) form fewer erythroid colonies than do cells cultured with human whole blood serum or fetal calf serum (P < 0.05). Treatment of washed platelets with thrombin releases a low molecular weight (<100,000) factor that enhances colony … Show more

“…PDGF also promotes in vitro multipotent haematopoietic progenitors and erythropoiesis. 23,24 In previous studies, we demonstrated the presence of functional PDGF receptors on human MK and MK cell lines 25 and showed that PDGF promoted MK colony formation. 26 In this study, we aimed to establish a cytokine combination for the efficient ex vivo expansion of the MK lineage from umbilical cord blood CD34 + cells, and to investigate the effect of PDGF in this system.…”

“…These CSFs include interleukin-3 (IL-3), granulocyte-macrophage CSF (GM-CSF) and granulocyte CSF (G-CSF) [6][7][8]. Also, platelet-derived growth factor (PDGF), activin, fibroblast growth factor (FGF) growth hormone, insulin and insulin-like growth factor-1 (IGF-1) augment erythropoiesis in vitro [9][10][11][12][13][14][15]. We identified an erythroid-potentiating activity (EPA) in the supernatant of the HTLV-II-transformed human T-lymphblast cell line (Me), and purified the 28 000 Da glycoprotein molecule to homogeneity [5,16].…”
