Platelet Concentrates Stored in Synthetic Medium after Filtration

Abstract: Platelet concentrates prepared from pooled buffy coats (BCPC) were stored in Plasma-Lyte A, a glucose-free synthetic medium, after leukocyte depletion by filtration through Pall PL 50, and compared to paired unfiltered BCPC stored in the same medium. Each pair of BCPC units was prepared from a pool of 10 buffy coats split into two indentical units. Platelet and leukocyte counts per unit of BCPC were 2.70 ±0.19 X 10^11 and 3.8 ±2.8x10^6 (filtered BCPC), 2.59 ±0.27 x 10^11 and 79 ±56x10^6 (control BCPC), respect… Show more

“…This is in accordance with Bertolini et al [12] who compared filtered and unfiltered platelet pools stored for 15 days in a synthetic medium. Other studies demonstrated that the in vitro and in vivo functional behavior of filtered platelets was satisfactory but no direct comparison was made with unfiltered platelets as was done in our study [13][14][15][16][17][18].…”

“…The other method is to sediment at high-Recently leukocyte depletion filters became available for PC. A few studies have shown the effectiveness of these filters [12][13][14][15][16][17][18], but little is known about the influence of fil tration on the quality of the platelets during subsequent stor age of the PC. We therefore evaluated 2 different leukocyte depletion filters for PC which have become available most recently: Pall PL50HF and Sepacell PL10-A.…”

“…Glycoprotein (GP) Ib and activation markers were evaluated as previously described [17] by flow cytometry using the followingmonoclonal antibodies: (1) CD42b (clone SZ2, Immunotech, Marseille, France) reactingwith GP Ib; (2) CD62 (clone CLB-thromb/6, Immunotech), reacting with alphagranule activation marker P-Selectin; (3) CD63 (clone CLB-gran 12, Immunotech), reacting with activation marker lysosome-like protein gp 53; (4) 9F9, reacting with plateletbound fibrinogen, a kind gift from Dr. A. Z. Budzynski (Philadelphia, Pa., USA); ( 5 ) D545, reacting withgranulophysin, an activation marker present on platelet dense granule membrane kindly provided by Dr. J.M.…”
