Platelet Aggregation Evoked In Vitro and In Vivo by Phosphatidic Acids and Lysoderivatives: Identity with Substances in Aged Serum (DAS)

Abstract: SummaryIn serum incubated at 36° C for 18-24 hours a factor (DAS) develops which on intravenous injection into cats evokes platelet aggregation followed by an increase in pulmonary vascular resistance (PVR). This change in PVR is mediated via the platelets since it significantly correlates with the preinjection platelet count. There is evidence that phosphatidic acids (PA) and lysophosphatidic acids (LPA) are the active components of DAS. Investigations performed on platelet-rich plasma from man, cat, pig, dog… Show more

“…2 Briefly, oleoyl chloride was acylated using 1-aminopropanol and pyridine followed by Dess-Martin oxidation. The resulting aldehyde was reacted with di-tbutylphosphonate in the presence of NaOH to generate the protected ␣-hydroxy derivative.…”

“…LPA has been shown to signal such vital cellular events as mitogenesis (1), platelet aggregation (2), tumor cell invasion (3), and escape from apoptosis (4). LPA is present in serum at low (1-20 M) micromolar concentrations (5) and is produced and released by stimulated platelets (6).…”

“…[7][8][9] In contrast, high micromolar concentrations of LPA are required to induce aggregation of platelet-rich plasma. [10][11][12] In the present study we investigated whether and through which mechanism LPA could induce platelet shape change and aggregation in whole blood. We also studied the formation of platelet-monocyte aggregates, an early marker of acute myocardial infarction.…”

“…It induced thromboembolism. 10 Interestingly, only human and cat platelets are sensitive to LPA. Platelets from other species such as pig, dog, rabbit, rat, and, very importantly, the mouse cannot be activated by LPA in vitro (Schumacher et al 10 and Tokumura et al 11 ).…”
