Platelet‐activating factor (PAF) induces activation of matrix metalloproteinase 2 activity and vascular endothelial cell invasion and migration

Abstract: Tumor-induced angiogenic responses lead to complex phenotypic changes in vascular endothelial cells, which must coordinate the expression of both proteases and protease inhibitors prior to the proliferation and invasion of surrounding stroma. Matrix metalloproteinase 2 (MMP2), which degrades Type IV collagen, is produced as proMMP2. proMMP2 is activated in part through its interactions with membrane Type 1 MMP (MT1-MMP) and tissue inhibitor of matrix metalloproteinase 2 (TIMP2). In this study, we demonstrate t… Show more

“…8, K and I), confirming our recent data that MMP2 mRNA levels are not affected by PAF (55). However, as expected, after 8 h of PAF stimulation, an elevated level of active MMP2 was observed in the medium, which increased rapidly at 16 and 24 h of PAF exposure (Fig.…”

“…Most recently, Axelrad et al (66) showed that PAF induced the transactivation of MT1-MMP and TIMP2 genes and the activation of MMP-2 in human umbilical vein endothelial cells. In these cells, MMP-2 activation occurred through a G␣ q protein-JAK-2-Src-mediated pathway (25,66).…”

In these cells, MMP-2 activation occurred through a G␣ q protein-JAK-2-Src-mediated pathway (25,66). Negative regulation of MMPs by PAF has also been reported; Barletta et al (67) showed that 300 nM PAF down-regulated MT1-MMP and MMP-2 mRNA levels and reduced MMP-2 activation in isolated human neuroblastoma cell clones.…”

“…The coordinate interaction of monocytes with activated platelets and collagen has been shown to induce the expression of MMP-9 (Galt et al, 2001). Likewise, proteolytic activity of endothelial cells is induced by their interaction with platelets (Axelrad et al, 2004). Therefore, as the role of MMPs in different biological systems is being revealed, there is a growing interest regard- ing the effect of platelets on protease production.…”
