Plasmid Profile and Antibiotic Resistance Pattern of Bacteria from Abattoirs in Port Harcourt City, Nigeria

Abstract: Several activities occur in abattoirs including receiving, slaughtering and sale of cattle for meat protein. In rearing of these animals, antibiotics and vaccines are incorporated as part of their meals as well as in the treatment of their infections. The regular use of antibiotics leading to the increased occurrence of antibiotic resistant organisms worldwide and also from abattoir samples, has prompted the determination of plasmid profile in these microorganisms as the plasmids act as a faster means of trans… Show more

“…Microorganisms isolated in this study were of the genus Staphylococcus, Pseudomonas, Klebsiella, Proteus and Bacillus. Similar organisms had been isolated from abattoir environments by some authors [36][37][38][39]. These microorganisms have been implicated as pathogens of various infections including diarrhoea, bacteremia, dysentery and urinary tract infections in humans which makes them of public health importance [40].…”

“…Drugs with lower Minimum Inhibitory Concentrations (MICs) such as Ofloxacin and Cloxacillin exhibited higher inhibitory activity against the isolates compared with those with higher MICs such as Augmentin which may be attributed to the frequent use of these antimicrobial drugs for treatment of infections; this trend has been reported by Adesoji et al [41] and Harrison and Bratcher [42], who studied the susceptibility of some microorganisms from abattoir sources to some drugs including Augmentin and cefuroxime. This poses a threat as treatment against infections caused by these organisms becomes difficult or may take a longer time to respond against the causative agent [39]. Microbial drug resistance has been reported to be linked with mechanisms such as inappropriate use of the drugs, widespread use of antibiotics in the production of animal feeds, its use in treatment of infections and for prophylaxis and excretion of metabolized or non-metabolized administered drugs by animals into the environment which undergo transformation into their active forms [43,19,44,45].…”

“…The presence of plasmid in Bacteria especially enteric organisms has been reported by other researchers in Nigeria [36] The presence of plasmid DNA in all the isolates implies that the resistance to commonly used antibiotics by this bacteria may be plasmid-mediated and also they serve as reservoirs for resistance plasmids that may be transferred to otherwise susceptible bacteria making them resistant, thus increasing the ocurrence of antibiotic resistance among microorganism. This in agreement with the finding of Ogbonna and Azuonwu [36].…”

“…Feacal Coliform counts was enumerated as described by Prescott et al [14]. Bacterial Colonies that appeared on the Erosin Methylene Blue (EMB) agar plates which were inoculated in duplicate with an aliquot of 0.1ml from 10 -2 dilutions and incubated 45.2 C for 24 hours, were counted and the mean expressed as cfu/ml for the samples [15]. While discrete colonies on the nutrient agar plates that were inoculated with an aliquot of 0.1ml from the direct samples were sub cultured on freshly prepared nutrient agar plate in order to isolate pure cultures.…”
