This article is available online at http://www.jlr.org functions, in addition to structural roles ( 1 ); consequently lipidomic studies have become fundamental for understanding their contribution to health and disease. Mass spectrometry (MS), with its capability of providing structural information, has been the main method of choice in lipidomic studies. Recent technological advances in mass spectrometers, including increased sensitivity, higher mass accuracy, higher scan speeds, and the ability to acquire in both positive and negative mode in one run have resulted in the increased popularity of MS as a detection technique for biomolecules in recent years. This has enabled the mapping of lipids present in fl uids and cells, leading to better understanding of the role of the different lipid classes in the pathophysiology of diseases.A critical step in lipidomic analysis is lipid extraction with an appropriate organic solvent mixture (solvent system) prior to MS detection. The solvent system should be capable of effectively extracting lipids representative of the sample under study without bias, inducing or promoting the degradation of lipids, or introducing contamination by nonlipid components such as sugars, peptides, and amino acids. Therefore, the success in the identifi cation and profi ling of lipids is critically dependent on the efficiency of the extraction step. Aim:The objective of the present study was to propose plasmalogens as a beneficial factor in human plasma by showing a highly positive correlation with high-density lipoprotein (HDL) and a significant reduction with aging. Methods: For 148 elderly subjects suspected of coronary artery disease (CAD), clinical characteristics such as coronary stenosis, hyperlipidemia, abnormal glucose tolerance, and hypertension were investigated, and serum biochemical markers including plasmalogens were determined. Results: Serum plasmalogens levels tended to fall in significant coronary stenosis and abnormal glucose tolerance. Correlative analyses among serum biochemical markers revealed that plasmalogens positively correlate with HDL-related values, particularly apolipoprotein A-(apo A-), and that the molar ratio of choline plasmalogen (ChoPlas) to ethanolamine plasmalogen (EtnPlas) correlates positively with low-density lipoprotein (LDL) particle size, and negatively with apo A-and fasting triglyceride (TG) levels. Comparison of plasmalogens in elderly subjects with those of 119 healthy young subjects showed a marked decrease in serum plasmalogens levels by aging. Conclusion: These results suggest that serum plasmalogens, antioxidant phospholipids, function as a beneficial factor as well as HDL, and that the measurement of serum plasmalogens is useful in clinical diagnosis.

The oxidative degradation of plasmalogen (alkenylacyl) phospholipids was analysed in the absence and the presence of polyunsaturated ester phospholipids by "H-NMR and by chemical determination. Brain lysoplasmenylethanolamine (lyso-P-PE), brain P-PE and erythrocyte P-PE, containing an increasing number of intrachain double bonds at sn # , were oxidized with 2,2h-azobis-(2-amidinopropane hydrochloride) (AAPH ; 2 or 10 mM) in Triton X-100 micelles (detergent\phospholipid 1 : 5, mol\mol). The formation of two peroxyl radicals was accompanied by the degradation of approx. one molecule of brain lyso-P-PE. On oxidation of brain P-PE or erythrocyte P-PE (320 nmol) with 2 mM AAPH, the (α-vinyl) methine "H signal of the enol ether decreased more rapidly than the methine proton peak of intrachain double bonds. The rate of enol ether degradation increased in the order : erythrocyte P-PE brain P-PE brain lyso-P-PE.
