Vitellogenin (vtg) was isolated from greenback flounder (Rhombosolea tapirina), rainbow trout, (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar) plasma. The molecular weight of each native molecule, as determined by gel filtration, was 540, 383 and 557 kD respectively. With SDS-PAGE under reducing conditions, rainbow trout vtg dissociated into 1 major subunit (~154 kD), and Atlantic salmon and greenback flounder vtg dissociated into 3 major subunits (~ 159, 117, 86 kD and 155, 104, 79 kD respectively). Polyclonal antisera, produced against only the highest molecular weight subunit from each species following excision of these bands from reducing gels, were reactive with all subunits in Western blots.These antisera were found to be highly species-specific as determined by competition ELISA.No cross-reactivity was noted, even between the two salmonid species. These data suggest that excision of bands from gels is a simple procedure for the preparation of highly specific antisera, and that cross-species assays employing heterologous antisera are unlikely to be possible.

Monocrotophos has been shown to exert estrogenic effects on teleosts. However, it has not been deter mined whether monocrotophos itself is the component that is responsible for exerting these estrogenic activities. Therefore, this study was carried out to investigate the estrogenic effects of monocrotophos standard in male goldfish (Carassius auratus), after a 21-day semi-static toxicity test. The induc tion of vitellogenin synthesis in males exposed to monocrotophos was confirmed with qualitative and quantitative methods. The observed increase in endogenous estrogen levels in males exposed to monocrotophos might explain the induction of vitellogenin. Moreover, we observed a decreased gonadosomatic index, ultrastructure damage of sperm and Sertoli cells, and several alterative testicular enzyme activities in males exposed to monocrotophos, which possibly affect spermatogenesis and male fertility. Our study showed the potential estrogenic effects of monocrotophos standard may act via the induction of vitellogenin synthesis and alteration of testicular ultrastructure and function in male goldfish.

A study was conducted to isolate, partial characterize Asian sea bass (Lates calcarifer) vitellogenin (vtg). Two-year-old juvenile L. calcarifer (n = 10) were given three intraperitoneal injections of 17-β estradiol (E2) at a dose of 2 mg/kg body weight to induce vitellogenesis. Blood was collected 3 days after the last injection, and plasma was purified through gel filtration chromatography. A broad single symmetrical peak consisting of vtg molecule was produced. Protein concentration was 0.059 mg/ml as determined by Bradfrod assay using bovine serum albumin as a standard. The protein appeared as one circulating form in Native PAGE considering the dimeric form of putative vtg with molecular weight of 545 kDa. In SDS-PAGE under reducing conditions, two major bands appeared at 232.86 and 118.80 kDa and minor bands at 100.60, 85.80 and 39.92 kDa, respectively. The purified vtg was used to generate a polyclonal antibody, and the specificity of antibody was assessed by Western blot analysis. Two major bands were immunoreacted, but no cross-reactivity was observed with plasma from non-induced males. The protein was characterized as phosphoglycolipoprotein as it positively stained for the presence of lipid, phosphorus and carbohydrate using Sudan Black B, methyl green and periodic acid/Schiff reagent solution, respectively. The amino acid composition was analyzed by high sensitivity amino acid analysis that showed high percentage of non-polar amino acids (~48 %). The results suggest the potential utilization of vtg as a basis tool to further study about reproductive physiology of this important economical species.
