Plasma MIC-1 and PAPP-A Levels Are Decreased among Women Presenting to an Early Pregnancy Assessment Unit, Have Fetal Viability Confirmed but Later Miscarry

Abstract: BackgroundWe have recently shown first trimester Macrophage inhibitory cytokine-1 (MIC-1) and Pregnancy Associated Plasma Protein-A (PAPP-A) serum concentrations are depressed among asymptomatic women destined to miscarry. Here we examined whether plasma levels of MIC-1 and PAPP-A are depressed among women presenting to an Early Pregnancy Assessment Unit (EPAU), noted to have a confirmed viable fetus, but subsequently miscarry.MethodsWe performed a prospective cohort study, recruiting 462 women in the first tr… Show more

“…GDF15 is a divergent member of the TGF-β superfamily ( 38 ). Its immunosuppressive function was first demonstrated in pregnancy, as GDF15 is highly expressed in the placenta during pregnancy and is essential to prevent rejection of the fetus ( 39 ). Zhou group reported that treatment with rhGDF15 in vitro prevents DC maturation and impairs the anti-tumor immune response of DCs ( 15 ).…”

“…We have previously shown [2] and then validated in two large prospective cohorts 3,5 that MIC-1 is depressed in viable pregnancies destined for miscarriage. Given the success in the strategy of focusing on an immune cytokine with a likely role in early pregnancy, attempts need to be made to identify and screen other such molecules for their predictive potential.…”

“…Subsequent studies showed that GDF-15 serum levels are increased in pregnant women at the onset of pregnancy and reach their highest concentration at the beginning of the third trimester (44). A retrospective study on sera collected during weeks 7-13 of pregnancy found comparatively lower GDF-15 serum levels in women who subsequently experienced miscarriages (45,46). Likewise, the observation that GDF-15 levels are elevated to a lesser extent in women with preeclampsia (with the more profound reduction found in late-onset cases) suggests that GDF-15 is a potential biomarker for tracking pregnancy and pregnancy-related complications (47).…”
