Plasma Metabolome Alterations Discriminate between COVID-19 and Non-COVID-19 Pneumonia

Abstract: Pneumonia is a common cause of morbidity and mortality and is most often caused by bacterial pathogens. COVID-19 is characterized by lung infection with potential progressive organ failure. The systemic consequences of both disease on the systemic blood metabolome are not fully understood. The aim of this study was to compare the blood metabolome of both diseases and we hypothesize that plasma metabolomics may help to identify the systemic effects of these diseases. Therefore, we profiled the plasma metabolome… Show more

“…To this point, a recent study did identify metabolites that were specific to acute respiratory distress syndrome patients where their disease was induced by COVID-19 compared to influenza [ 40 ]. Other studies report oral metabolites that differentiated patients with COVID-19 from those with influenza [ 41 ], and plasma metabolites that distinguish between COVID-19 and non-COVID-19 pneumonia [ 42 , 43 ]. We do emphasize, though, that in this study, care was taken to account for basic demographics, risk factors, and comorbidities.…”

“…Extraction was performed as previously described [96,97]. Briefly, 20 µL plasma sample was added to 180 µL ice-cold methanol:H 2 O (8:1) containing 2 µg/mL D6-pentanedioic acid (C/D/N Isotopes, Quebec, Canada) as internal standard.…”

“…Metabolite extraction was performed as per our previous publication ( 21 ). Prior to extraction, the serum samples were thawed on ice for 30 minutes.…”
