Plasma glycomics predict cardiovascular disease in patients with ART‐controlled HIV infections

Abstract: Despite effective control of HIV infection with antiretroviral drugs, individuals with HIV have high incidences of secondary diseases. These sequelae, such as cardiovascular disease (CVD), are poorly understood and represent a major health burden. To date, predictive biomarkers of HIV-associated secondary disease have been elusive, making preventative clinical management essentially impossible. Here, we applied a newly developed and easy to deploy, multitarget, and high-throughput glycomic analysis to banked H… Show more

“…Before analyzing the IgG itself, we also confirmed that the HcKO and BHcKO mice lacked ST6Gal1 activity in the plasma using a plate assay as we have previously described (27). We found that both the WT and BcKO strains showed strong ST6Gal1 activity in plasma samples when supplied with exogenous CMP-sialic acid, but neither the HcKO nor BHcKO mice showed detectable activity (Fig 5A).…”

“…High throughput, multiplexed lectin ELISA was performed on whole plasma samples, as has been published (27,34). Briefly, plasma was diluted to 0.5 μg/ml in carbonate coating buffer (100 mM NaHCO 3 , 30 mM NaCO 3 , pH 9.5), pipetted into a 384 well black ELISA plate, and incubated overnight at 4 °C.…”

“…Receiver Operating Characteristics (ROC) analysis, a well-established machine learning technique generating a model able to distinguish between different classes, already proved to be a valuable tool with enormous clinical potential in diagnosing several pathologies [ 51 , 52 , 53 , 54 ]. Nevertheless, this methodology is usually applied to samples obtained with invasive techniques, such as intravenous blood collection or biopsy.…”

“…1), whether the HIV-induced changes in the circulating glycome are linked to inflammaging and HIV-associated co-morbidities (during both viremic and ART-suppressed HIV infection) is less clear. Recently, plasma glycomic biomarkers were identified (using lectins) to predict HIV-associated cardiovascular events [95]. However, more work is needed in this direction.…”
