Plasma endocannabinoids and cannabimimetic fatty acid derivatives are altered in gastroparesis: A sex‐ and subtype‐dependent observation

Abstract: Background: Gastroparesis (GP) is a motility disorder of the stomach presenting with upper gastrointestinal symptoms in the setting of delayed gastric emptying. Endocannabinoids are involved in the regulation of GI function including motility. However, their role in the pathophysiology of GP has not been sufficiently investigated. Our goal was to compare the circulating levels of endocannabinoids and cannabimimetic fatty acid derivatives in GP versus control subjects. Methods: The study compared plasma concent… Show more

“…These techniques can be applied to different samples (plasma, brain regions, skeletal muscle, adipose tissues, etc.). For example, using this technique, AEA, palmitoyl-ethanolamide (PEA), oleoylethanolamide (OEA), 2-AG and its isomer 1-AG [78], and N-acyl-ethanolamines (NAEs) and 2-monoacylglycerols (2-MAGs), congeners of AEA and 2-AG [82,83], have been determined in plasma. Similarly, eCBs and their derivatives have been measured in rat liver tissue [84] and AEA metabolite formation in kidney tissue samples [85] by HPLC-MS/MS.…”

“…eCB ligands and related endolipids are not limited to the brain, and from a translational standpoint measuring endolipids in biological specimens that are less invasive to sample, such as plasma, could be particularly informative. Already, we have shown sex differences in endolipids in plasma of patients with gastroparesis 22 . Likewise, serum collected from cannabis users shows increased palmitic acid, oleic acid, 2-AG, and PEA, but not AEA or OEA compared to non-cannabis users 23 , which illustrates that plasma endolipids are responsive to changes in systemic CB pharmacology.…”

“…11,12 These enzymes are also involved in the processing of a wide range of endogenous lipids with cannabimimeticlike properties; however, there are limited available data on their physiological roles in the development and progression of human disorders, including DGBI. 13 Given the importance of the eCB system in the pathophysiology of CVS and their role as a modulator of the autonomic nervous system, which is dysregulated in CVS, we tested the hypothesis that CVS is associated with changes in circulating eCB and related lipid signaling molecules, and these changes are potentiated by the cephalic-vagal stimulation during sham feeding.…”
