Plasma clearance of rat bikunin: evidence for receptor-mediated uptake

Abstract: Bikunin is a chondroitin sulphate-containing protease inhibitor with a molecular mass of 25 kDa. It is secreted into the blood by hepatocytes, and recent observations indicate that it may have an extravascular function. Here we have studied the plasma clearance of bikunin in rats and mice. On intravenous injection, radiolabelled bikunin was found to have a half-life of 10 min; in rats with ligated renal arteries, the clearance time was twice as long, implying that the kidneys account for half the uptake. As ju… Show more

“…Free BK is cleared through the kidneys by glomerular filtration (35,36) and then partly reabsorbed and catabolized by proximal tubular cells. In our study, increased BK excretion in urine during inflammation was not related to any modification in its behavior in the kidney.…”

“…This was, however, not included in the calculation of θ for bikunin. It has previously been determined in mice that bikunin does not form aggregates or split products in plasma (Sjoberg et al. 1995).…”

“…Thus, it is still possible to compare the t 1/2 of sL-selectin with that of other plasma proteins. Small molecular mass proteins such as IL-2 and bikunin have reported t 1/2 of 4 and 10 min, respectively, due to rapid kidney clearance (53,54). Because sL-selectin is a relatively high molecular mass protein, it would not be expected to be removed to any significant amount by filtration in the kidney.…”
