ABB), a unique local banana cultivar from Pasuruan, East Java, Indonesia. Biodiversitas 23: 3521-3532. Characterization of local banana (Musa spp.) germplasm is fundamental for identification and provides information on traits that support their sustainable conservation and optimum utilization. This study aims to characterize and discuss the potential and conservation of Pisang Kates,a local cultivar originatingfrom Tutur, Pasuruan, East Java, Indonesia. The observed characteristics include morphology, molecular, agronomy and yield performance, leaf anatomy and stomata, pollen morphology and viability, fruit total soluble solids and nutrients. Results showed that Pisang Kates hasa unique fruit shape ovoid to globose, weight of40-150 g, appear solitary and randomly attached directly to peduncle, like papaya. It was molecularly by Internal Transcribed Spacer barcode confirmed as ABB genotype. It is tolerant to drought and banana bunchy top virus, but susceptible to wilts. Leaf anatomy has three palisades and isodiametric polygonal epidermals. Leaf stomata is amphistomatic, tetracytic on adaxial,and paracytic on abaxial. Pollen unit is monad, very large, subspheroidal, monocolpate aperture, granulum ornamentation, and low viability. Fruit moderately sweet with nutrient contents equal to dessert banana. Pisang Kates has potential as one serving consumer size fruit, ornamental fruit plant, female parent for breeding,and pioneer plant for rehabilitation programs. Both ex-situ and on-farm conservation strategies of this valuable local banana genetic resource are encouraged.

Black Zira (Bunium persicum) is an important endemic medicinal plant in Iran. This research was conducted out to assay breaking seed dormancy, develop new protocols for callus induction, somatic embryogenesis, and arti cial seed. Seeds were treated with ultrasonic waves in different periods (0, 1, 3, 5, 7, and 9 min). Then, two experiments with IBA (0, 1, 2, 3, and 4 mg L − 1 ) and a combination of different levels of 2,4-D (0, 1, 2, and 4 mg/l) and BAP (0, 0.5, 1, and 2 mg/l) were designed. Different concentrations of sodium alginate (2, 3, and 4%) and CaCl 2 (50, 75, and 100 mM) were prepared for arti cial seed production. According to the results, the 7 min ultrasonic waves had the maximum effect on germination (81.87%), while the highest negative effect on germination (2.5%) was reported from 9 min exposure time. In the IBA experiment, the highest percentages of callogenesis, callus weight, and adventitious root were obtained from MS medium containing 4 mg/l IBA. Moreover, 1 mg/l IBA resulted in the highest embryogenesis percentage. In the second experiment, the highest percentage of callus formation and callus weight were achieved in 1 mg/l BAP + 4 mg/l 2,4-D, 0.5 mg/l BAP + 4 mg/l 2,4-D, and 2 mg/l 2,4-D media. The maximum embryogenesis percentage was seen in 0.5 mg/l BAP + 1 mg/l 2,4-D, 0.5 mg/l BAP + 2mg/l 2,4-D, and 1 mg/l 2,4-D media. The highest germination rate (77.77%) was obtained from encapsulated embryos with 3% sodium alginate and 100 mM CaCl 2 . Key MessageA protocol with improved results has been developed for indirect somatic embryogenesis and, arti cial seed production in B.persicum using hypocotyl explants.

The aim of this study was to check the use of the following gelling agents: the xanthan gum “Adicel®” and the stabilizer “Super Liga Neutra®” to replace agar in the in vitro rooting phase of Gerbera hybrida cv. Essandre. Additionally, the possibility of using chemical sterilization of both culture media and glassware with sodium hypochlorite to replace autoclaving was analyzed. The gelling agents, xanthan gum “Adicel®” and the stabilizer “Super Liga Neutra®” were tested at the following concentrations (g L-1): 7; 9; 11; 13; 15; 17; 19; and 21. No concentrations of Super Liga Neutra® provided effective solidification. Concentrations of 17 and 21 g of Adicel® provided a good gelling of the culture medium, which was compared to the medium containing agar (control) with two types of sterilization: autoclaving (for 20 and 40 minutes) and chemical sterilization. Autoclaving for 20 minutes did not provide effective elimination of contamination in the culture medium containing xanthan gum; this only occurred when autoclaving time increased to 40 minutes. Plant development in culture media containing 17 and 21 g of xanthan gum, either sterilized by autoclaving for 40 minutes or at a concentration of 17 g xanthan gum using sodium hypochlorite, was statistically the same as the control that contained agar. However, plant development at a concentration of 21g of xanthan gum in a sterilized medium using sodium hypochlorite was lower than that observed in media containing agar.
