The purpose of this study was to develop a new micropropagation system for Tylophora indica, an important medicinal plant in India, using root explants as starting material. Root explants cultured on MS medium supplemented with 6-benzyladenine (BA) or 2-isopentyladenine (2iP) produced organogenic nodular meristemoids (NMs) within 4 weeks. NMs induced from the cut ends of root segments showed two types of organogenic response--direct shoot bud formation and somatic embryogenesis--when maintained on induction medium. In 42% of the explants, NMs developed shoot buds directly in the presence of 10.72-26.80 microM BA. On average, 18.5 +/- 0.7 shoots per gram of NM tissue were obtained after each 4-week subculture. Elongation of microshoots and root initiation were correlated with the auxin used, with the optimal response occurring in the presence of 28.54 microM indole-3-acetic acid. In 39% of the explants, NMs dedifferentiated into friable embryogenic callus (FEC) in the presence of BA or 2iP after 12 weeks of culture. Of the different treatments, MS medium supplemented with 10.72 microM BA was the most effective in inducing FEC and somatic embryogenesis: at this concentration 64% of the cultured NMs developed FEC and, on the same medium, 89% of the FEC produced globular somatic embryos (SEs). FEC biomass increased nearly five-fold with every 4-week subculture, and about 30 SEs were recovered per gram of FEC during this period. The best conversion of mature SEs to complete plantlets was obtained on basal MS medium-42%. Plantlets derived via somatic embryogenesis and shoot organogenesis were successfully hardened (88-96%) and transferred to the field.

Plants of two cytotypes (2n=2x=20, and 2n=3x=30) of pinto peanut (Arachis pintoi Krapov. & W.C. Gregory) were regenerated through somatic embryogenesis. Embryogenic calli were induced from shoot tips or immature leaves dissected from in vitro growing plants. In the case of the diploid peanut the best somatic embryogenesis was achieved when shoot tips were cultured on Murashige and Skoog (MS) medium supplemented with 10 mg dm -3 Picloram (PIC) and 0.1 mg dm -3 6-benzylaminopurine (BAP) or when explants from immature leaves were cultured on MS + 10 mg dm -3 PIC + 0.01 mg dm -3 BAP. In the case of triploid peanut the highest number of somatic embryos was obtained when shoot tips were cultured on MS + 10 mg dm -3 PIC + 0.01 mg dm -3 BAP or when immature leaves were cultured on MS + 20 mg dm -3 PIC + 0.01 mg dm -3 BAP. Somatic embryos were converted into plants by culture on MS + 0.01 mg dm -3 naphthaleneacetic acid + 0.01 mg dm -3 BAP. Plants were successfully transferred to pots in greenhouse.

The study presents the in vitro potential of preconditioned plumular apices explants of peanut for shoot regeneration. Mature embryo explants were isolated and preconditioned for 10 days on MS medium with 10 and 20 mg/l BAP (6-Benzylaminopurine) followed by postconditioning of isolated plumular apices postconditioned on MS medium enriched with 0.25-2.0 mg/l BAP singly or with 0.25 mg/l NAA (1-Naphthaleneacetic acid). Preconditioning of explants using 20 mg/l BAP was found to be more regenerative to induce 4.38 shoots with shorter shoots (0.75 cm) compared to 3.50 shoots and shoot length of 1.25 cm when preconditioned with 20 mg/l BAP. Number of shoots per explants ranged 2.30-5.36 and 3.17-6.0 respectively on explants preconditioned with 10 and 20 mg/l BAP and postconditioned on 0.25-2.0 mg/l BAP -0, 0.25 mg/l NAA with shoot length in range of 1.01-2.30 cm and 0.74-1.80 cm, respectively. Maximum number of shoots (6.0) with stunted shoots (0.74 cm) were recorded from 20 mg/l preconditioning followed by culture on medium containing provided with 1.0 mg/l BAP+ 0.25 mg/l NAA. Irrespective of preconditioning treatments, minimum number of shoots per explants but relatively longer shoots were scored on MS medium provided with 0.25 mg/l BAP+0.25 mg/l NAA. Regenerated shoots were successfully rooted using 0.25-1.0 mg/l IBA. They were transferred to pots containing peat moss that resulted in successful acclimatization of in vitro regenerated plantlets at room temperature.
