Plant polyphenols mobilize endogenous copper in human peripheral lymphocytes leading to oxidative DNA breakage: A putative mechanism for anticancer properties

Abstract: Plant polyphenols are important components of human diet and a number of them are considered to possess chemopreventive and therapeutic properties against cancer. They are recognized as naturally occurring antioxidants but also act as prooxidants catalyzing DNA degradation in the presence of transition metal ions such as copper. Using human peripheral lymphocytes and Comet assay we have previously confirmed that resveratrol-Cu(II) is indeed capable of causing DNA degradation in cells. In this paper we show tha… Show more

“…The involvement of ROS has been proposed as upstream event occurring during treatment with high (micro-molar) concentrations of trans-resveratrol [1,21], as well as with other polyphenols, including epigallocatechin-3-gallate [44,53), woodfordin I [35] and quercetin [33,64]. In our experimental system ROS burst occurs precociously (within the first hour) and the detrimental effects produced were confirmed by the increase in lipid peroxidation by-products, MDA and 4-HNE.…”

“…25 These data are confirmed by many authors using doses comparable to those used in our article. 26 The pro-oxidant action which involves the mobilization of endogenous copper could be a common mechanism for anticancer and chemopreventive properties of plant polyphenols. These results are also consistent with what is well known about other polyphenols which have both anti-and pro-oxidant activity 27 and inhibit topoisomerase II activity depending on concentrations, duration and mode of treatment, cell types.…”

“…In addition to their antioxidant properties, it is worthy to note that cytotoxic effects of RES, PIC, and TPS have also been reported. In this regard, RES has demonstrated pro-oxidant, genitourinary, immunotoxic, hepatotoxic and genotoxic properties [18][19][20][21][22][23]. PIC, a metabolite of RES, has been reported to induce apoptosis of transformed and primary lymphocytes [17] and to also exhibit cytotoxic actions towards macrophages in vitro [24].…”
