The introduction of dwarfing genes triggered a wave of “green revolution”. A number of wheats dwarfing genes have been reported in previous studies, and only a small fraction of these have been applied to production practices. Therefore, the development of novel dwarfing genes for wheat is of great value. In this study, a novel dwarfing site, Rht-yz, identified in the Yanzhan mutation, is located on chromosome 4B (30-33MB) and its mechanism of action is different from that of Rht-B1b (C-T mutation), but whether it affects the Rht-B1a (TraesCS4B02G043100) or other genes is unclear. Exogenously applied GA3 experiments showed that Rht-yz is one of the gibberellin-insensitive dwarf genes. The effects of the dwarf gene Rht-yz on agronomic traits in wheat were evaluated in the field using Yanzhan, Yanzhan mutations, F2:3 and F3:4 lines. The results showed that Rht-yz improved lodging resistance by reducing plant height, increasing diameter, wall thickness and mechanical strength of the basal stem. In terms of yield traits, Rht-yz had negative effects on tiller number plant-1, biomass plant-1 and yield plant-1, but had no significant effect on harvest index, 1000-kernel weight and spike traits. In addition, Rht-yz significantly increased crude protein, wet gluten and starch content. Therefore, the rational use of the new dwarfing site Rht-yz has potential and value in dwarf wheat breeding.

As a new synthetic species, triticale has been widely used for food and feed. Lodging is a severe problem in triticale production, and it is unknown whether wheat height‐reducing genes can improve lodging resistance in triticale. In this study, the wheat‐dwarfing genes Rht12 and Rht‐B1b were introgressed into hexaploid triticale (AABBRR) lines to understand their effect on plant height. The results showed that Rht12 and Rht‐B1b reduced the plant height of triticale plants by 37.7 and 29.9%, respectively. Rht12 and Rht‐B1b both had significant effects on the basal internode's length, diameter, and thickness, which may greatly improve lodging resistance. Additionally, the net photosynthetic rate was increased by 28.7% in Rht12 dwarf lines. However, Rht12 had a significant and negative effect on thousand‐kernel weight (14.5%) and yield per plant (20.6%). On the other hand, Rht‐B1b reduced the number of tillers by 19.6% and yield per plant by 18.1% with no negative effect on the harvest index. Furthermore, Rht12 and Rht‐B1b decreased grain plumpness but improved several grain quality traits. Conclusively, Rht12 and Rht‐B1b improved lodging resistance but had some negative effects on yield components, which need careful consideration in triticale breeding.

Preparation of comb‐type grafted hydrogels composed of polyacrylamide and chitosan and their use for DNA adsorption

Comb-type grafted hydrogels composed of polyacrylamide (PAAm) and chitosan (CT) were prepared and used for DNA adsorption. Instead of direct grafting of the acrylamide monomer onto the CT chain, semitelechelic PAAm with carboxylic acid end groups (PAAm-COOH) was synthesized by free-radical polymerization with mercaptoacetic acid as the chain-transfer agent, and it was grafted onto CT with amino groups. The synthesis of telechelic PAAm-COOH and the formation of comb-type grafted hydrogels were confirmed by attenuated total reflectance/Fourier transform infrared spectroscopy and scanning electron microscopy measurements. The prepared comb-type grafted hydrogels were used as sorbents in DNA adsorption experiments conducted at þ4 C in a tris(hydroxymethyl)aminomethane/ ethylenediaminetetraacetic acid solution of pH 7.4. DNA adsorption capacities as high as 2.0 Â 10 3 lg of DNA/g of dry gel could be achieved by the comb-type hydrogels with higher PAAm contents. This value was approximately 6 times higher than that of CT alone. In addition, the comb-type hydrogels showed a high adsorption/desorption rate depending on the PAAm content in the hydrogel. As a result, these comb-type hydrogels carrying higher amounts of DNA may be considered good candidates for achieving higher removal rates for anti-DNA antibodies and for effective gene therapy systems.
