Plant Expression of a Bacterial Cytochrome P450 That Catalyzes Activation of a Sulfonylurea Pro-Herbicide

Abstract: The Sfreptomyces griseolus gene encoding herbicide-metabolizing cytochrome P450sul (CYP105Al) was expressed in transgenic tobacco (Nicofiana tabacum). Because this P450 can be reduced by plant chloroplast ferredoxin in vitro, chloroplast-targeted and nontargeted expression were compared. Whereas P450sul antigen was found in the transgenic plants regardless of the targeting, only those with chloroplast-directed enzyme performed P450sul-mediated N-dealkylation of the sulfonylurea 2-methylethyl-2,3-dihydro-N-[(4,… Show more

“…The cytochrome P 450 negative selection marker was subcloned as a PstI fragment from plasmid pSSU-SU121 (O'Keefe et al, 1994) into pBluescript KS. The linker oligo 5¢-CTAGACCATGGCTTC-CTCTGTGATTTCCAGTGCA-3¢ restored the sequence of the petunia RBCS transit peptide while introducing an NcoI site on the ATG start codon.…”

“…The transposase gene fragment, coding for both the TnpA and TnpD proteins, was cloned under the control of a 35S promoter, the Spm promoter, or a meiosis-specific promoter from the AtDMC1 gene (constructs 8313, 8337, and 8353, respectively; see Methods; see also Klimyuk and Jones, 1997). The SU1 gene from Streptomyces griseolus encodes a cytochrome P450 that confers sensitivity to R7402, which is a sulfonylurea proherbicide from Du Pont (O'Keefe et al, 1994). The counterselection cassette with the SU1 gene was cloned between the left border and the transposase gene in the same orientation.…”

“…Chloroplast thylakoid membranes contain ferredoxin (Fd) and ferredoxin:NADP ϩ reductase (FNR), which serve as electron carriers in photosynthetic electron transport chain to produce NADPH. By a reverse reaction, these proteins can also serve as electron donors from NADPH for various physiological enzymes (for review, see Knaff and Hirasawa, 1991;Neuhaus and Emes, 2000), for chloroplast-targeted bacterial P450 in vivo (O'Keefe et al, 1994), and for the reconstituted in vitro P450 system (Yamazaki et al, 1995;Dong et al, 1996). However, the NADPH-FNR-Fd electron transport chain is operative in the stroma and is not likely to support P450s in the chloroplast outer membrane.…”
