PlGFenhancesTLR‐dependent inflammatory responses in human mononuclear phagocytes

Abstract: Problem Levels of placental growth factor (PlGF) peak during third trimester of pregnancy, a time when women are at increased risk of virus-induced morbidity. We hypothesized PlGF might contribute to an exaggerated inflammatory response to toll-like receptor (TLR)-activation. Method of Study Primary human adult and cord blood CD14+ cells were cultured in the presence of TLR-ligands and/or PlGF. Results PlGF significantly enhanced the magnitude and duration of TNF mRNA and protein production by TLR-7/8 acti… Show more

“…PlGF significantly increases IL-6 and IL-8 secretion, cyclooxygenase-2 (Cox-2) expression, and consequent prostaglandin (PG)-E2 and PG-F2α release, and MMP-9 gene expression and enzyme production. [106,145]. Accordingly, PlGF significantly enhanced the magnitude and duration of TNF-α mRNA and protein production by TLR-7/8-activated monocytes and increased the subsequent production of TNF-α-independent inflammatory cytokines.…”

“…PlGF was recently found to enhance the inflammatory response of monocytes upon TLR7/8 activation, TLRs typically activated by single stranded RNA viruses. 25 The low PlGF in RVU may represent a mechanism to hamper excessive inflammation to control ocular damage, which is in agreement with the immune privileged site of the eye. Our study includes measurements of intraocular fluids without simultaneous analyses of cytokines in serum.…”

“…It has been supported that there is a close correlation between the dysregulation of IL-6 with moderate and severe BPD in preterm infants with a small gestational age [35]. PlGF overexpression contributes an exaggerated in ammatory state [36]. MiR-214 can inhibit angiogenesis procession by suppressing Quaking and pro-angiogenic growth factor expression [37].…”
