PKC Activation Induces Inflammatory Response and Cell Death in Human Bronchial Epithelial Cells

Abstract: A variety of airborne pathogens can induce inflammatory responses in airway epithelial cells, which is a crucial component of host defence. However, excessive inflammatory responses and chronic inflammation also contribute to different diseases of the respiratory system. We hypothesized that the activation of protein kinase C (PKC) is one of the essential mechanisms of inflammatory response in airway epithelial cells. In the present study, we stimulated human bronchial lung epithelial (BEAS-2B) cells with the … Show more

“…Verification of siRNA knockdown was performed as described previously (36). Western blotting was performed according to a standard protocol (37). Briefly, cells were lysed with radioimmunoprecipitation assay (RIPA) buffer (50 mM Tris, 150 mM NaCl, 0.1% SDS, 0.5% sodium deoxycholate, and 1% Triton X-100).…”

“…Secretory factors in the culture medium were detected via MILLIPLEX MAP Human Cytokine/Chemokine-Premixed 42 Plex (Millipore; Billerica, MA) (18). Concentrations of IL-6 (DY206), IL-8 (DY208), and GRO-1a (DY275) were measured with DuoSet 1 enzyme-linked immunosorbent assay (ELISA) systems (R&D Systems, Minneapolis, MN).…”

“…Furthermore, thymol significantly inhibited the phosphorylation of PKCα and ERK1/2, as well as the activation and expression of MMP9 and MMP2. PKC is an intracellular signaling protein of the protein kinase family that regulates cell survival, proliferation and differentiation, and the cell cycle (25)(26)(27). PKC phosphorylation is involved in malignant transformation and tumor promotion, as well as in invasion and metastasis.…”
