Pituitary-testicular function is suppressed by an LHRH antagonist but not by an LHRH agonist in the marmoset monkey

Abstract: The use of pituitary desensitization by an LHRH agonist (buserelin) to examine pituitary-testicular function was investigated in a New World primate. Six adult male marmoset monkeys were injected s.c. with an LHRH agonist implant (1.5 mg in a rod 0.5 cm long). Pharmacokinetics, determined by radioimmunoassay of urinary buserelin, revealed a rapid initial release of the agonist followed by a steady decline during a 200-day period. The LHRH agonist treatment resulted in a rapid initial rise in plasma LH followed… Show more

“…In these studies, neither local testosterone nor supplemented human chorionic gonadotropin (hCG) could induce maturation of the immature testicular tissues of marmoset in nude mice host. Moreover, increased levels of gonadotropin after gonadectomy (Hodges 1978), but failure of gonadotrophin-releasing hormone (GnRH) agonist to stimulate gonadotropin and testosterone concentrations (Lunn et al 1990) further suggest the peculiar endocrine regulation of testicular function in this species as demonstrated previously by us , Müller et al 2004.…”

“…The assay sensitivity was 2-5 pmol/tube and inter-and intra-assay coefficients of variation were 11-1 and 4-2% respectively. Testosterone After extraction, plasma testosterone was measured by RIA as described by Lunn et al (1990). Intra-and interassay coefficients of variation were 4-5 and 11 0% respectively, the assay sensitivity being 50nmol/1.…”

“…Assay data were corrected to allow for a mean recovery after extraction of 68%. Minor modifications from the procedure used by Lunn et al (1990) involved the use of an increased plasma volume for extraction (from 10 to 20 pi), all of the extracted plasma samples subsequently being measured in triplicate, rather than in duplicate.…”

“…The marmoset (Callithrix jacchus) is an animal model for many reproductive biology studies and has been used extensively to study the effects of GnRH agonists and antagonists in vivo (Lunn et al 1990). The marmoset, unlike most other species, exhibits maintained secretion of luteinising hormone (LH) and testosterone after treatment with GnRH agonist implants, but not after GnRH antagonist treatment.…”
