Piroxicam and intracavitary platinum-based chemotherapy for the treatment of advanced mesothelioma in pets: preliminary observations

Abstract: Malignant Mesothelioma is an uncommon and very aggressive tumor that accounts for 1% of all the deaths secondary to malignancy in humans. Interestingly, this neoplasm has been occasionally described in companion animals as well. Aim of this study was the preclinical evaluation of the combination of piroxicam with platinum-based intracavitary chemotherapy in pets. Three companion animals have been treated in a three years period with this combination. Diagnosis was obtained by ultrasonographic exam of the body … Show more

“…Our data support previous observations [ 30 , 42 , 43 ] of a synergistic effect of piroxicam, when used in combination with CDDP on cell cycle regulation and apoptosis. Interestingly, the specific check-points affected by this treatment are not overlapping in different cell lines, this demonstrating that the effects of piroxicam could be on multiple targets.…”

“…The results were confirmed in vivo , in a mouse MM model indicating that piroxicam and cisplatin association specifically acts on cell cycle regulation triggering apoptosis, and may hold promise in the treatment of MM [11]. Finally in spontaneous MM in pets, we recently have been able to show that piroxicam/cisplatin combination has remarkable efficacy at controlling the malignant effusion secondary to MM in our samples [18].…”

“…Intravenous carboplatin use in cats is less frequently reported; however, reported uses include extraskeletal osteosarcoma (Spugnini and others 2001, Dhaliwal and others 2003) and a variety of sarcomas and carcinomas (Kisseberth and others 2008). Carboplatin has also been reported to be administered by intracavitary injection for mesothelioma in dogs and cats (Spugnini and others 2008) and intralesional injection for feline squamous cell carcinoma (Theon and others 1996).…”
