Oral lyophilisates are one of the orodispersible tablets produced by lyophilization technique. Due to their porous structure, they instantly disintegrate when contacting saliva. Generally, they are suitable for drugs with a short onset of action, such as nonsteroidal anti-inflammatory drugs (NSAIDs). Therefore, formulations of piroxicam lyophilisates and their properties were investigated. Because piroxicam is classified as a low solubility but high permeability drug, a solubility enhancing agent should be included in the formulation. The influence of β-cyclodextrin (βCD) and hydroxypropyl-β-cyclodextrin (HPβCD) on disintegration time and morphology of piroxicam lyophilisates was studied. Furthermore, the satisfactory taste-masking efficiency of HPβCD in piroxicam lyophilisates was studied by sensory test. The effect of matrix polymer mixtures on disintegration time was previously evaluated. The appropriate ratio of polymer mixture, i.e., gelatin and hydroxypropyl methylcellulose (HPMC), provided lyophilisates with the fastest disintegration time (<30 s) was designated for further studies. The presence of both CDs decreased disintegration time and modified the surface and matrix of lyophilisates. With the theoretical study based on molecular mechanics methods and the investigation through the chemical shift of protons using nuclear magnetic resonance (NMR), piroxicam formed inclusion complexes with both βCD and HPβCD. Dissolution profiles of piroxicam/HPβCD lyophilisates revealed improved piroxicam solubility by inclusion complex formation. Finally, the piroxicam/HPβCD lyophilisates demonstrate the bitterness suppression in healthy volunteers.

In this ever-growing world, it is crucial to improve upon the formulations in terms of potency, patient acceptability, fewer side effects, and quicker relief. Due to these requirements, the market is flooded with various combination dosage forms, with a constant increase in number. Paracetamol is a commonly used non-steroidal anti-inflammatory drug (NSAID) that has antipyretic and analgesic action. This drug is available in a wide range of combinations. It acts by inhibiting the production of prostaglandins, which combat pain and inflammation. A simultaneous multicomponent analysis is used to determine the estimation of medicines that are available in combination. Different analytical techniques are available for their determination, one of which includes the use of UV spectrophotometric methods. This review focuses on a variety of paracetamol combinations with drugs like Domperidone, Aceclofenac, Diclofenac Sodium, Etodolac, Ibuprofen, Piroxicam, Caffeine, Aspirin, and their simultaneous estimation by different UV methods viz. Simultaneous equation method, Absorbance ratio (Q-Analysis), Difference spectrophotometry, Derivative spectroscopy method, and a few other chemometric methods. This manuscript would provide the platform to have exhaustive literature on methods used for the estimation of paracetamol with different drugs using a spectrophotometer. It would help the researchers and scholars who are working in the area.

Repurposing of Drug Candidates for Treatment of Skin Cancer

Skin cancers are highly prevalent malignancies that affect millions of people worldwide. These include melanomas and nonmelanoma skin cancers. Melanomas are among the most dangerous cancers, while nonmelanoma skin cancers generally exhibit a more benign clinical pattern; however, they may sometimes be aggressive and metastatic. Melanomas typically appear in body regions exposed to the sun, although they may also appear in areas that do not usually get sun exposure. Thus, their development is multifactorial, comprising endogenous and exogenous risk factors. The management of skin cancer depends on the type; it is usually based on surgery, chemotherapy, immunotherapy, and targeted therapy. In this respect, oncological treatments have demonstrated some progress in the last years; however, current therapies still present various disadvantages such as little cell specificity, recurrent relapses, high toxicity, and increased costs. Furthermore, the pursuit of novel medications is expensive, and the authorization for their clinical utilization may take 10–15 years. Thus, repositioning of drugs previously approved and utilized for other diseases has emerged as an excellent alternative. In this mini-review, we aimed to provide an updated overview of drugs’ repurposing to treat skin cancer and discuss future perspectives.
