“…Piperazic acid is one of the most common N-N containing building blocks of natural products, being found in numerous natural compounds with activities ranging from antibiotics to immunosuppressants. All piperazic acid-containing molecules known to date come from Actinomycete bacteria isolated from diverse environments (Morgan et al 2019 ), but the structural and functional variety is remarkable: kutznerides, padanamides, himastatins, monamycines, polyoxypeptin and sanglifehrins, to name a few, contain piperazic acid (Fig. 7a ).…”

“…The biosynthetic origins of piperazate-containing natural products is a young area of study 19 and our findings have important implications for biochemists studying the incorporation of these unusual amino acids into secondary metabolites. Our proposed scaffold would be only the second class of natural products bound at N1 of the piperazate (the first being the sanglifehrins) and the only class to be synthesized entirely on an NRPS assembly line.…”

“…Natural products with nitrogen–nitrogen (N−N) bonds are a group of rarely isolated yet highly bioactive molecules that includes drugs and drug candidates . Unlike many natural products, the biosynthetic routes to molecules containing N−N bonds remained enigmatic until recently .…”
