Pioglitazone Inhibits Diabetes-Induced Atrial Mitochondrial Oxidative Stress and Improves Mitochondrial Biogenesis, Dynamics, and Function Through the PPAR-γ/PGC-1α Signaling Pathway

Abstract: Background: Oxidative stress contributes to adverse atrial remodeling in diabetes mellitus. This remodeling can be prevented by the PPAR-γ agonist pioglitazone via its antioxidant and anti-inflammatory effects. In this study, we examined the molecular mechanisms underlying the protective effects of pioglitazone on atrial remodeling in a rabbit model of diabetes. Methods: Rabbits were randomly divided into control, diabetic, and pioglitazone-treated diabetic groups. Echocardiographic, hemodynamic, and electrophy… Show more

“…Pioglitazone is a synthetic PPARγ agonist and is the first-line drug for the clinical treatment of type 2 diabetes ( Zhang et al, 2021 ). It exerts its pharmacological effects by activating PPARγ in the liver and fat cells.…”

“…Furthermore, the PPAR signaling pathway also plays a key role in mitochondrial function. In the diabetes-induced atrial, the agonist of PPAR-g significantly increases mitochondrial biogenesis-related transcription factors (PGC1a, NRF1-2, TFAM) and mtDNA copy number and reduces mitochondrial ROS production (66).…”

“…PGZ acts as a PPAR agonist, known to increased PGC1α activity and mitochondrial biogenesis [ 136 ]. However, more recently, PGZ was shown to protect the expression of fusion (OPA1, MFN1/2) and fission (DRP1) proteins in a rabbit model of diabetes-induced atrial remodelling [ 137 ]. PGZ had similar effects in trisomic fetal fibroblasts where OPA1 and MFN1 gene expression was restored and DRP1 inhibited [ 138 ].…”
