Pilot study on peptide purity—synthetic oxytocin

Abstract: Under the auspices of the Protein Analysis Working Group (PAWG) of the Comité Consultatif pour la Quantité de Matière (CCQM) a pilot study, CCQM-P55.2.b, was coordinated by the Bureau International des Poids et Mesures (BIPM) and the Chinese National Institute of Metrology (NIM). Three Metrology Institutes or Designated Institutes and the BIPM participated. Participants were required to assign the mass fraction of oxytocin (OXT) present as the main component in the comparison sample for CCQM-P55.2.b. The compa… Show more

“…In its programmes related to metrology for health, comparison reports on oxytocin (a peptide calibrator material) coordinated by the BIPM (CCQM-K155.b [32] and -P55.2.b [33]) were published. A paper describing methods of impurity quantification in oxytocin standards was submitted for publication.…”

“…Due to the molecular properties of the intact analytes, the qNMR of protein is challenging. Efforts to quantify peptides of a limited length and individual amino acids have been made [ 22 , 23 , 24 , 25 , 26 ]. Digestion of intact protein into individual amino acids provides an avenue for qNMR analysis; cleaving a protein into individual amino acids enhances the signal-to-noise ratio by the respective number of the respective amino acid by making these amino acids chemically equivalent.…”
