Pillararene‐Based Supramolecular Vesicles for Stimuli‐Responsive Drug Delivery

Abstract: Supramolecular vesicles (SMVs) self‐assembled from the supra‐amphiphiles, consisting of two scaffolds linked together through noncovalent interactions, can realize stimuli‐responsive controlled release of encapsulated drugs for enhanced therapeutic efficacy and minimized side effect of drugs. Pillararenes (PAs), an emerging kind of macrocyclic hosts in 2008, are easy to modify with a variety of functionalities. SMVs from PAs and specific guests mainly based on the host–guest interactions have attracted increas… Show more

“…1D] [145] . These hydroquinone units are generally 5-10, with P[5]A and P [6]A being the most common [146][147][148][149][150][151] . P[n]As are widely used in various fields such as drug delivery, ion recognition, adsorptive separation, sensors, and optoelectronic materials due to the characteristics of symmetrical rigid skeleton, adjustable electron-rich cavity, and easy functionalization [152][153][154] .…”

“…Supramolecular chemistry provides a wide range of receptor molecules, particularly macrocycles, that can be used as hosts in a variety of applications, garnering considerable interest from the scientific community. In terms of structural diversity, macrocycles, include cyclodextrins, , cucurbiturils, , crown ethers, cryptands, pillararenes, and calixarenes, , to give some examples. On the other hand, guest molecules are generally small molecules or ions that benefit from the host–guest interactions to improve their stability and bioavailability or even to tune their spectroscopic features, namely, colorimetric and fluorescent properties.…”

“…Among these macrocyclic molecules, pillararenes (P[n]A) have attracted wide interest owing to their symmetrical structure and easy modication. [50][51][52][53][54][55][56][57][58] The electron-rich inner cavity of pillararenes (P[n]A) gave them a high host-guest binding ability, thus providing them with excellent loading potential. 59 Therefore, the construction of a pillararene-based supramolecular nanoplatform by host-guest interaction would provide a versatile and promising strategy to intelligently combine multimodal therapies for efficient cancer treatment.…”
