PIE-1 is a bifunctional protein that regulates maternal and zygotic gene expression in the embryonic germ line of Caenorhabditis elegans

Abstract: Histone deacetylation plays an important role in methylated DNA silencing. Recent studies indicated that the methyl-CpG-binding protein, MBD2, is a component of the MeCP1 histone deacetylase complex. Interestingly, MBD2 is able to recruit the nucleosome remodeling and histone deacetylase, NuRD, to methylated DNA in vitro. To understand the relationship between the MeCP1 complex and the NuRD complex, we purified the MeCP1 complex to homogeneity and found that it contains 10 major polypeptides including MBD2 and… Show more

“…For germline development, pie-1 and nos-2 control PGC fate and influence their incorporation into the gonadal primordium (Seydoux et al, 1996;Subramaniam and Seydoux, 1999;Tenenhaus et al, 2001). In this paper, we report that the C. elegans Hand bHLH transcription factor hnd-1 is important for early gonadogenesis as well as for embryogenesis.…”

“…OsDOS encodes a CCCH-type zinc finger protein without known signal peptide. Apparently, unlike the cytoplasm localization of Pie-1, Mex-1, and Pos-1, three Caenorhabditis elegans proteins with similar zinc fingers (Guedes and Priess, 1997; Tabara et al, 1999;Tenenhaus et al, 2001), OsDOS was found to be a nuclear-localized protein (Fig. 1B) similar to HUA1, a nuclear-localized CCCH-type zinc finger protein in Arabidopsis (Li et al, 2001).…”

“…We then investigated whether polarity cues regulate PLK-1 asymmetry separately from asymmetric spindle positioning and cell fate determination, both of which are also controlled by A-P polarity (reviewed by Gönczy and Rose, 2005). To this end, we examined goa-1/gpa-16(RNAi) embryos, in which the first division is equal as a result of symmetric spindle positioning (Colombo et al, 2003;Gotta and Ahringer, 2001), as well as pie-1(zu154) mutant embryos, in which the fate of the P 1 blastomere is altered (Tenenhaus et al, 1998;Tenenhaus et al, 2001). We found that PLK-1 distribution is asymmetric in such embryos (Fig.…”
