Piceatannol ameliorates behavioural, biochemical and histological aspects in cisplatin‐induced peripheral neuropathy in rats

Abstract: Peripheral neurotoxicity is a dose-limiting and a potentially lifelong persistent toxicity of cisplatin. This study investigated the possible protective effect of piceatannol (PIC) in a model of cisplatin-induced peripheral neuropathy in rats. PIC (10 mg/kg, i.p.) was given for 7 days, starting 2 days before cisplatin single injection (7 mg/kg, i.p.). Behavioural, biochemical and histological examinations were conducted. Cisplatin administration resulted in thermal hypoalgesia evidenced by increased paw and ta… Show more

“…dose of CIS disrupted neuromuscular coordination, resulted in muscle weakness, and decreased latency to fall in the hanging wire “grip” test. Our results run in accordance with Shabani et al ( 2012a , b ) and Wahdan et al ( 2021 ). This might be ascribed to the neurotoxic potential of CIS to disrupt the neuronal function in the cerebellar cortex, especially Purkinje cells which are the essential part for movement control and motor functions (Pisu et al 2004 ; Avella et al 2006 ).…”

“…In addition, CIS administration provoked thermal hyperalgesia and stimulated sensitivity to pain, this runs in accordance with several studies (Ali et al 2014 ; Sharawy et al 2015 ; Ahmad et al 2017 ; Saadati et al 2021 ; Wahdan et al 2021 ). CIS-induced thermal hyperalgesia might be attributed to the axonal deterioration in the sciatic nerve and the lower spine of the spinal cord, indicating the potential of CIS to affect small fibers that are closely involved in thermal hypersensitivity (Liu et al 2022 ).…”

“…Several cytotoxic compounds including cisplatin may facilitate impairment of LTP. Cisplatin-induced cognitive impairment was documented in animals models using Morris water maze [ 33 , 34 , 35 , 36 , 37 , 38 , 39 , 40 ], Y-maze test [ 33 , 41 , 42 , 43 , 44 ], T-maze [ 45 , 46 , 47 ], and novel object recognition test [ 9 , 48 , 49 , 50 , 51 ], including in humans in the treatment of oncological diseases using chemotherapy using cisplatin as a chemostatic [ 12 , 52 , 53 , 54 , 55 ].…”
