PI3K is negatively regulated by PIK3IP1, a novel p110 interacting protein

Abstract: Signaling initiated by Class Ia phosphatidylinositol-3-kinases (PI3Ks) is essential for cell proliferation and survival. We discovered a novel protein we call PI3K Interacting Protein 1 (PIK3IP1) that shares homology with the p85 regulatory PI3K subunit. Using a variety of in vitro and cell based assays, we demonstrate that PIK3IP1 directly binds to the p110 catalytic subunit and modulates PI3K activity. Our studies suggest that PIK3IP1 is a new type of PI3K regulator.

“…PIK3IP1 is a relatively recently identified PI3K regulating protein. PIK3IP1 binds directly to PI3K and negatively regulates its activity (Zhu et al, 2007). PI3K pathway activation is seen in many types of cancers, and the pathway regulates several cellular processes that are critical for cancer progression, including metabolism, growth and survival (Courtney et al, 2010).…”

“…Vock et al ( 27,28 ) examined 0.2 mM oleate for 24 h and 50 M palmitate. They identifi ed subunit of PI3 kinase ( 26 ), suggesting that downregulation of PIK3IP1 might mediate the protective effects of oleate. We fi rst tested the effects of the pharmacologic PI3 kinase inhibitor LY294002 in cells treated with palmitate (0.5 mM) or palmitate and oleate (0.5 mM and 0.25 mM, respectively).…”

“…In the current study, oleate adaptation completely abolished the PIK3IP1 transcript response to palmitate in HepG2 cells. PIK3IP1 was recently identifi ed by Zhu et al ( 26 ) as a protein that shows homology to the regulatory p85 subunit of PI3K. PIK3IP1 binds to and negatively regulates the p110 subunit of PI3K, thus acting in place of the p85 regulatory subunit.…”

“…His 6 -tagged PR8/NS1 was generated by partially digesting pGEX'TEV'PR8/NS1 with NcoI/NotI and ligating the ϳ700-bp fragment into the corresponding sites of linearized pEHISTEV. The pGADT7.p110␣-ABD plasmid (expressing HA-tagged p110␣-ABD) was a kind gift from M. DeFrances (University of Pittsburgh) (27). The integrity of all newly generated constructs was confirmed by DNA sequencing.…”
