PI3K/Akt/mTOR signaling pathway and targeted therapy for glioblastoma

Abstract: Glioblastoma multiform (GBM) is the most common malignant glioma of all the brain tumors and currently effective treatment options are still lacking. GBM is frequently accompanied with overexpression and/or mutation of epidermal growth factor receptor (EGFR), which subsequently leads to activation of many downstream signal pathways such as phosphatidylinositol 3-kinase (PI3K)/Akt/rapamycin-sensitive mTOR-complex (mTOR) pathway. Here we explored the reason why inhibition of the pathway may serve as a compelling… Show more

“…Yuge et al [36] demonstrated that nicotine promoted tumorigenesis and induced chemoresistance by activating the PI3K/Akt/mTOR pathway in bladder carcinomas . Furthermore, Li et al [37] verified that the inhibition of the PI3K/Akt/mTOR pathway could be a promising targeted therapy for glioblastoma, which was the most malignant type of gliomas ; this finding was in accordance with our results.…”

“…For its elusive response to standard treatments, the development of novel therapeutic approaches for GBM remains indeed a pivotal step in the modern oncologic research. Protocols based on radionuclide therapy appear to be particularly promising for the GBM treatment [2,[4][5][6][7]. Experimental evidence has clearly demonstrated that cancer cells have a particularly high fractional content of copper inside the cytoplasmic and nuclear fraction compared to normal cells.…”
