Oxidative stress is a condition that is characterized by the generation of reactive oxygen species more than the amount endogenous antioxidants can absorb. Several diseases are associated with oxidative stress, notably during disease progression. Thus, the present study aimed to determine the phytoconstituents and antioxidant potential of the ethyl acetate leaf extract of Corchorus olitorius (ELEC). The phytochemicals were qualitatively and quantitatively determined, followed by characterization using Fourier-transform infrared (FTIR) spectroscopy. The antioxidant potential was determined in vitro. Alkaloids, saponins, and flavonoids were detected in concentrations of 8.50 ±2.65, 11.83 ±0.73, and 19.17% ±0.73, respectively. The FTIR spectrum revealed 9 peaks including six at the group frequency region corresponding to alcohols, carboxylic acid, amine salts, alkenes, alkyne, esters, and oxime functional groups. The ELEC exhibited lower total reducing power (11.06 ±1.34 AAE µg/ml) compared to its total antioxidant capacity (49.26 ±2.44 AAE µg/ml) with a higher (60.47% ±2.44) percentage inhibition of peroxidation than AA (37.98% ±1.88). Furthermore, the ELEC exhibited a lower (0.20 ±0.01 nmol/ml) MDA concentration than AA (0.42 ±0.02 nmol/ml). Conclusively, C. olitorius might be applied for the management of oxidative stress-linked ailments and a source of novel therapeutics for these ailments.

The novel coronavirus of 2019 (nCoV-19) has become a pandemic, affecting over 205 nations with over 7,410,000 confirmed cases which has resulted to over 418,000 deaths worldwide. This study aimed to identify potential therapeutic compounds and phytochemicals of medicinal plants that have potential to modulate the expression network of genes that are involve in SARS-CoV-2 pathology in human host and to understand the dynamics key proteins involved in the virus-host interactions. The method used include gene network analysis, molecular docking, and sequence and structure dynamics simulations. The results identified DNA-dependent protein kinase (DNA-PK) and Protein kinase CK2 as key players in SARS-CoV-2 lifecycle. Among the predicted drugs compounds, clemizole, monorden, spironolactone and tanespimycin showed high binding energies; among the studied repurposing compounds, remdesivir, simeprevir and valinomycin showed high binding energies; among the predicted acidic compounds, acetylursolic acid and hardwickiic acid gave high binding energies; while among the studied anthraquinones and glycosides compounds, ellagitannin and friedelanone showed high binding energies against 3-Chymotrypsin-like protease (3CL pro), Papain-like protease (PL pro), helicase (nsp13), RNA-dependent RNA polymerase (nsp12), 2'-O-ribose methyltransferase (nsp16) of SARS-CoV-2 and DNA-PK and CK2alpha in human. The order of affinity for CoV proteins is 5Y3E > 6NUS > 6JYT > 2XYR > 3VB6. Finally, medicinal plants with phytochemicals such as caffeine, ellagic acid, quercetin and their derivatives could possibly remediate COVID-19.

The aim of the study was extraction of arial part of Euphorbia cyathophora constituents with methanol and evaluate its effect on mitotic index and total chromosomal aberration bone marrow cell and spleen cell in mice 200 gm of E. cyathophora fine powder was defatted then extracted by cold maceration 80% ethanol for seven days. The extract was filtered and dried in a rotary evaporator then the dried extract was suspended with water and consecutively extracted using chloroform, ethyl acetate for each. The aqueous layer was then mixed with 100ml methanol. These fractions are dried under reduced pressure to obtain the dry extract. Twenty-four Albino mice were used for the experiment. The animals were divided into four groups: Group 1: Mice were treated with distilled water. The dose was given daily for seven successive days. Group 2: Mice were treated with a single dose (20mg/kg) of methotrexate (positive control). Group 3: Mice were treated with (100mg/kg) of menthol fraction for seven successive days. Group 4: Mice were treated with (200mg/kg) of methanol fraction for seven successive days. Mice were sacrificed by (spinal dislocation). Samples of bone marrow cells and spleen cells were taken and genotoxic analyses. Methanol fraction of Euphorbia cyathophora at a dose of 100mg/kg demonstrated a significant decrease in mitotic index and a significant increase in total chromosomal aberrations as compared to distilled water in both bone marrow cells and spleen cells (p<0.05). 100 mg/kg and 200 mg/kg of methanol fraction of Euphorbia cyathophora that showed to be significantly higher in mitotic index and significantly lower in total chromosomal aberration as compared to methotrexate (p<0.05). In conclusion the current study revealed that the methanol fraction of aerial parts of Euphorbia cyathophora is genotoxic but its genotoxicity is less than that of methotrexate.
