The seed extracts of Nigella sativa (black cumin) have been used in different civilizations around the world for centuries to treat various animal and human ailments. C. albicans is a member of the normal human microbiome, but under certain circumstances it can cause infections that range from superficial infections of the skin to life-threatening systemic infections. Resistance to antifungal drugs in C. albicans has been on the increase in recent years. Several factors such as biofilm formation and overexpression of genes for efflux drug pump have been identified to contribute to the pathogenic potential of this fungus. This research examined antifungal effects of black cumin seed extracts and seed oil on C albicans wild-type (C1 and C2) and URA3 delete (CAF2 and CAI4) strains. Effects of the extracts on the key genes (MDS3 and MDR1) involved in drug resistance in C. albicans were also investigated. Aqueous and methanol extracts of the plant seeds were prepared using standard procedure. Aqueous and methanol extracts of N. sativa seeds, as well as the seed oil, were used for antimicrobial screening and for the analysis of effects on drug resistance genes using agar well diffusion technique, while fluconazole was used as a standard drug. The seed oil, as well as the aqueous and methanolic seed extracts of N. sativa greatly inhibited the wild-type clinical isolates C1 and C2. Interestingly, the URA3-delete strains were only inhibited by the oil. The amplification of the target genes in the various strains presented some intriguing results, with the gene(s) detected only prior to exposure or after exposure to the agent or at both instances. N. sativa, particularly the seed oil, has proven to have antifungal effects on fluconazole-resistant C. albicans with varying effects on the resistant genes.

Sweet orange fruit (Citrus sinensis,L.) peels were dried to constant weight using oven, box solar and sun drying methods. The dried orange fruit peels were processed into flours and used to substitute 10, 20 and 30 % wheat flour (WF) in biscuits. The orange peel flours were analyzed for photochemical composition. The biscuits were analyzed for photochemical composition and evaluated for sensory properties. The content of flavanoids (0.48 mg/100g) of the solar dried orange fruit peel flour (SOFPF), saponins (0.73 mg/100g), tannins (0.91 mg/100g) and alkaloids (0.99 mg/100g) of oven dried orange fruit peel flour (ODOFPF) were higher than those of the sun dried orange fruit peel flour (SDOFPF). The carotenoids (0.32 mg/100g), flavanoids (0.38 mg/100g) of 30 % SDOFPF biscuit, saponins (0.33 mg/100g), tannins (0.59 mg/100g) and alkaloids (0.82 mg/100g) of the 30 % ODOFPF based biscuit were higher than those of the other biscuits. The 100 % WF biscuit (control), followed by 10 % SDOFPF biscuit had higher overall acceptability scores of 8.0 and 7.1, respectively than the other biscuits. The overall acceptability score of 5.5 for biscuit containing 30 % ODOFPF was the lowest.

The aim was to extract bioactive compounds from the husk and pulp of Dioscorea bulbifera L. by the combination of ultrasound and solid-liquid extraction techniques, as well as to evaluate the antioxidant potential. The extraction at 80°C for 60 minutes showed the highest content of total phenolic compounds (TPC), 715.53 ± 8.00 mg EAG 100 g− 1, for the husk, while the extraction at 70°C for 45 minutes showed 235.50 ± 25.30 mg EAG 100 g− 1, for the pulp. The extracts with the highest TPC were evaluated for the other bioactive compounds. The husk and pulp extracts showed flavonoid content of 363.63 ± 8.92 and 102.44 ± 1.51 mg EC 100 g− 1, respectively. The estimation of the total carotenoid content allowed obtaining 2.13 ± 0.11 µg 100 g− 1 for the husk and 1.34 ± 0.11 µg 100 g− 1 for the pulp. The antioxidant potential was evaluated according to the FRAP assay (125.09 ± 8.52 and 32.76 ± 0.65 µM ferrous sulfate g− 1), the removal of H2O2 (29% and 41%), the ABTS radical assay (66.88 ± 0.93 and 14.93 ± 0.31 µM Trolox g− 1) and the β-carotene/linoleic acid system, (84% and 47%) for husk and pulp, respectively. The bioaccessibility was obtained around 25% (pulp) and 16% (husk) accessible for absorption in the intestine. The combination of ultrasound and solid-liquid extraction methods proved to be effective in extracting bioactive compounds, which is a potential source for isolation and purification of bioactive compounds, with possible applications in the food industry.
