Physiology of IgD III. Effect of treatment with anti‐IgD from birth on the magnitude and isotype distribution of the immune response in the spleen

Abstract: Continued treatment with monoclonal anti-IgD (Ig-5a) from birth in BALB/c mice causes a markedly increased responsiveness to i.v. injected dinitrophenylated ovalbumin (DNP-OVA) with Bordetella pertussis at the age of 8 weeks. The 19S plaque-forming cell (PFC)/spleen response is particularly enhanced, 6-8-fold, but all the other isotypes also show increases of 2-6-fold, including IgA and IgE. Both primary and secondary PFC responses and serum antibody titers are enhanced. After transfer of spleen cells from ant… Show more

“…Previous studies of the splenic B cell population of mice treated chronically from birth with anti4 antibody have demonstrated that these cells share some phenotypic and functional characteristics with splenic B cells from neonatal mice [l, 3, [12,37,391, low-angle light scatter analyses of the donor cells which home to the recipient spleens showed that these cells not only failed to express increased sIa densities, but actually exhibited reduced numbers of sIa molecules (M.…”

“…For example, it has been shown that treatment of mice from birth with allogeneic anti-IgD suppresses cell-surface IgD expression and down-regulates the production of antibodies of all isotypes to antigens administered s.c. or i.p. (4,5,8). However, the greatly reduced numbers of remaining 4+, 5-B cells in such mice were shown to be actually hyperresponsive to antigens injected i.v.…”

“…The significance of surface IgD in regulation has been demonstrated in many investigations (4)(5)(6)(7)(8)(9)(10). For example, it has been shown that treatment of mice from birth with allogeneic anti-IgD suppresses cell-surface IgD expression and down-regulates the production of antibodies of all isotypes to antigens administered s.c. or i.p.…”
