Physicochemical Properties of Collagen from Acaudina Molpadioides and Its Protective Effects against H2O2-Induced Injury in RAW264.7 Cells

Abstract: Collagen is a promising biomaterial used in the beauty and biomedical industries. In this study, the physicochemical characterization, antioxidant activities, and protective effects against H2O2-induced injury of collagen isolated from Acaudina molpadioides were investigated. The amino acid composition analysis showed that the collagen was rich in glycine (Gly), alanine (Ala), and glutamic acid (Glu), but poor in tyrosine (Tyr) and phenylalanine (Phe). Zeta potential analysis revealed that the isoelectric poin… Show more

“…On the contrary, the contents of positively charged amino acids (histidine [His] and lysine [Lys]) and hydrophobic amino acids (methionine [Met], tyrosine [Tyr], and phenylalanine [Phe]) were very low. The amino acid compositions of AMP were consistent with those of collagen obtained from A. molpadioides (J. Li et al., 2020). It was reported that Met, Tyr, His, and Cys (cysteine) could enhance the antioxidant activities of peptides (Ao & Li, 2012).…”

“…Collagen of A. molpadioides was obtained according to the method reported in our previous study (J. Li et al., 2020). The collagen was added into a buffer solution (pH = 10.0, w/v = 1/100) and pretreated for 30 min under microwave irradiation of 250 W. The mixtures were hydrolyzed at 45°C for 1 h by alkaline protease (≥200 U/mg).…”

“…GSH-Px converts reduced glutathione to oxidized glutathione while reducing lipid hydroperoxides to their corresponding alcohols or free hydrogen peroxide to water [25]. Figure 5b shows that the GSH-Px activity in the H 2 O 2 group was significantly lower than that of the blank control group but increased significantly after pretreatment with those peptide mixtures.…”
