Physicochemical characterization of a PEGylated liposomal drug formulation using capillary electrophoresis

Abstract: In this work, the applicability of using CE to perform a physicochemical characterization of a PEGylated liposomal drug formulation of the anti-cancer agent oxaliplatin was examined. Characterization of the liposomal drug formulation using CE instrumentation encompassed: determination of the electrophoretic mobilities, size determination by Taylor dispersion analysis and interaction studies. Electrophoretic mobilities determined by CE were compared with the results obtained by laser Doppler electrophoresis, wh… Show more

“…Therefore, the potential therapeutic application of a given liposome is highly influenced by its average size and, consequently, by the possible liposome-cell interactions at the target sites of action. Table 2 lists some techniques used to physically characterize the liposomes, in terms of the size distribution and zeta field-flow fractionation [68 -70] zeta potential LDE capillary electrophoresis [68,71] rsif.royalsocietypublishing.org J. “…The ease of use of DLS and its wide availability made it a popular technique for particle sizing, however the presence of large aggregates or dust, which scatter more than the small particles, tends to bias the measured average hydrodynamic radius toward larger size, mainly at low scattering angles (Cottet et al, 2007;Fischer and Schmidt, 2016;Franzen et al, 2011). Another widely used technique, particularly in biological applications, is the fluorescence correlation spectroscopy (FCS).…”

“…The increased viscosity decreases the mobility of the charged analytes. As a result, the mobility of Apep10 was corrected according to Franzen et al [26]. The electropherograms of Apep10 and acetone in the running buffer with various concentrations of the E .…”
