Physical Proximity and Functional Interplay of the Glycoprotein Ib-IX-V Complex and the Fc Receptor FcγRIIA on the Platelet Plasma Membrane

Abstract: Although the glycoprotein (GP) Ib-IX-V complex andFc␥RIIA are distinct platelet membrane receptors, previous studies have suggested that these structures may be co-localized. To determine more directly the proximity of GP Ib-IX-V and Fc␥RIIA, we assessed the effects of anti-GP Ib␣ monoclonal antibodies on Fc␥RIIA-mediated platelet aggregation and on the direct binding of polymeric IgG to human platelets. In addition, we directly examined the proximity of Fc␥RII and GP Ib-IX-V using flow cytometric fluorescence… Show more

“…In this respect, it has been reported that vWF-induced platelet activation requires activation of the Fc receptor ␥II (or Fc receptor ␥-chain)-protein-tyrosine kinase Syk-signaling pathway ( Fig. 7) (43)(44)(45)(46). In addition, GPIb-IX is associated with several intracellular signaling proteins including 14-3-3 (47,48), phosphatidylinositol 3-kinase (via 14-3-3) (49), and calmodulin (50).…”

“…As the only IgG FcR expressed by human platelets, FcγRIIA contributes to the pathophysiology of diseases such as heparininduced and antibody-mediated thrombocytopenias and to antiphospholipid antibody syndrome-mediated arterial thrombosis. In addition, FcγRIIA has been shown to stimulate platelets through interaction with other molecules and receptors, including α2bβ3, GP1b-IX-V and von Willebrand factor [25][26][27]. Furthermore, FcγRIIA and PECAM-1 are physically and functionally associated on the surface of human platelets [28].…”

“…29 GPVI also relies on an immunoreceptor tyrosine-based activation motif on the cytoplasmic domain of associated FcR ␥-chains for maximal signal transduction. Further studies involving the relationship between these platelet receptors are warranted.…”
