Physical map of the Treponema denticola circular chromosome

Abstract: A physical map of the Treponema denticola ATCC 33520 genome was constructed by pulsed-field gel electrophoresis and DNA hybridization. The organism possesses a single, circular chromosome of approximately 3.0 Mbp and a 2.6-kbp circular plasmid, pTD1. The physical map of the A؉T-rich genome was constructed with the rare-cutting restriction enzymes AscI, NotI, and SrfI, which have 8-bp G؉C-rich recognition sites. The genes flgE, tdpA, and prtB encoding the flagellar hook protein, a 53-kDa immunogenic protein, an… Show more

“…As a positive control, the genome size of T. denticola ATCC 33520 was determined. The result obtained, 2,984 kb, agrees closely with that previously obtained, 3,030 kb (5). The genome sizes for seven species representing six distinct groups of treponemes are surprisingly consistent at 2.2–2.5 Mbp.…”

“…The purpose of this work was to determine the genome sizes of representative strains of the cultivable oral treponeme groups using pulsed‐field gel electrophoresis. This approach has been used to accurately estimate the size of the genome of T. denticola ATCC 33520 prior to its sequencing (5). Such information allows inferences to be made about the complexity of the genome of each Treponema group.…”

“…It has been proposed that the 1.14 Mb T. pallidum genome was derived from the genome of T. denticola or other host-associated treponemes through deletion and divergence (15). Comparisons reveal that the T. pallidum genome shares limited nucleotide identity with the T. denticola genome.…”

“…Moreover, genome sequencing has revealed considerably larger genomes of oral treponemes (e.g. 2.84 Mb for T. denticola , (MacDougall and Girons, 1995; Seshadri et al, 2004) compared to T. pallidum strains (1.14 Mb; Fraser et al, 1998). In this review, we will focus on the genetic diversity of obligatory pathogenic Treponema pallidum strains and on related uncultivable treponemes.…”
