Physical and transcriptional organisation of the bread wheat intracellular immune receptor repertoire

Abstract: 1Disease resistance genes encoding intracellular immune receptors of the nucleotide-binding 2 and leucine-rich repeat (NLR) class of proteins detect pathogens by the presence of pathogen 3 effectors. Plant genomes typically contain hundreds of NLR encoding genes. The availability 4 of the hexaploid wheat cultivar Chinese Spring reference genome now allows a detailed study 5 of its NLR complement. However, low NLR expression as well as high intra-family sequence 6 homology hinders their accurate gene annotation… Show more

“…We annotated 792 full-length rye NLR genes (tbls. S-NLR—S-RLOC), finding them enriched in distal chromosomal regions, similar to what has been seen recently in the bread wheat genome 9,63 (fig. M-GENESa; Note S-NLR).…”

“…At this stage, we expect that deep next‐generation sequencing approaches or whole genome sequencing can shed some light into this issue. Alternatively, if the goal is the detection of new functional genes with resistance activity against biotrophic fungi, RenSeq approaches (Steuernagel et al , ) targeting sequences such as nucleotide‐binding site–leucine‐rich repeats (NB‐LRRs) with known participation in plant resistance should be also appropriate.…”

“…NLR gene signatures were detected with NLR-Annotator [82] using a sequence fragment length of 20 kb with 5 kb overlaps. NLR-Annotator predictions for Nipponbare were compared to previously annotated NLR genes using BED-tools intercept [83].…”
