Abstract. Previous studies have demonstrated the ultrasound-induced skin optical clearing enhancement with topical application of 60% glycerol ͑G͒ on in vitro porcine skin and in vivo human skin. Our purpose was to find the relation between the effect of optical skin clearing and different concentrations of glycerol and to find more effective ultrasound-glycerol combinations on optical skin clearing. The enhancement effect of ultrasound ͓Sonophoresis ͑SP͒ delivery͔ in combination with 40% G, 60% G, and 80% G on in vitro human skin optical clearing was investigated. Light imaging depths of skin were measured using optical coherence tomography. Different concentrations of glycerol and ultrasound with a frequency of 1 MHz and an intensity of 0.5 W / cm 2 was simultaneously applied for 15 min. The results show that with the increase of concentration of glycerol, the optical clearing of skin is much improved. Optical clearing capability of glycerol was more enhanced with simultaneous application of ultrasound compared with glycerol alone. The attenuation coefficients of skin tissues after application of 40% G/SP, 60% G/SP, and 80% G/SP decreased approximately 11.8%, 18.5%, and 20.0% at 15 min compared with 40% G, 60% G, and 80% G alone, respectively. The greatest decrease in attenuation coefficients at 60 min was approximately 52.3% and 63.4% for 80% G ͑without ultrasound͒ and 80% G/SP ͑with ultrasound͒, respectively, which are 2.1-fold and 2.6-fold to that in the 40% G.

ABSTRACT. The correlation between skin barrier function and transepidermal water loss (TEWL) was evaluated in dogs. Stratum corneum (SC) of 10 healthy dogs was removed by tape stripping (TS), which decreased the corneal layer to allow for permeation of fluorescent dye into skin. TEWL of damaged skin was measured with the closed-chamber-type TEWL analyzer, CC-01. The frequency of TS was directly related to the decrease of SC and the increased permeation of fluorescent dye, and TEWL increased with increasing impairment of skin barrier function. Thus, TEWL is thought to be a useful parameter that characterizes skin barrier function in human.Many skin disorders are suggested to be associated with skin barrier function in dogs [7]. However, the relationship between skin disease and the barrier function is not well analyzed because of paucity of studies on the TEWL value in normal and damaged canine skin. In this study, we evaluated whether TEWL could reflect impaired canine skin barrier function or not.Ten healthy dogs (8 crossbred dogs and 2 beagles, age range 7-13 years, 1 female, 3 sterilized females, and 4 neutered males) were enrolled in this study. All animals were fed on the same food and kept in an air-conditioned room. Two different anatomical areas, lumbar skin and inguinal skin, were used for TEWL measurement. Hair on both areas of each dog was clipped before measurement. Measurements were performed at 23-25°C and 45-65% relative humidity. All dogs were acclimated at least 30 min to the measurement conditions. Abstract. FDA's critical path initiative documents have focused on the challenges involved in the development of new drugs. Some of the focus areas identified apply equally to the production of generic drugs. However, there are scientific challenges unique to the development of generic drugs as well. In May 2007, FDA released a document "Critical Path Opportunities for Generic Drugs" that identified some of the specific challenges in the development of generic drugs. The key steps in generic product development are usually characterization of the reference product, design of a pharmaceutically equivalent and bioequivalent product, design of a consistent manufacturing process and conduct of the pivotal bioequivalence study. There are several areas of opportunity where scientific progress could accelerate the development and approval of generic products and expand the range of products for which generic versions are available, while maintaining high standards for quality, safety, and efficacy. These areas include the use of quality by design to develop bioequivalent products, more efficient bioequivalence methods for systemically acting drugs (expansion of BCS waivers, highly variable drugs), and development of new bioequivalence methods for locally acting drugs.

Abstract: The tapes utilized removed a similar amount of SC. The tapes have a different propensity to cause barrier disruption. Some individuals do not demonstrate increased TEWL despite an equivalent mass of SC being removed compared to those who do show a response.
