Physical and Chemical Adsorption of Mucor javanicus Lipase on SBA-15 Mesoporous Silica. Synthesis, Structural Characterization, and Activity Performance

Abstract: In this work a sample of SBA-15 mesoporous silica was synthesized and characterized by TEM, XRD, and N2 adsorption. The sample had a high value of specific surface area (1007 m2 g(-1)) and total pore volume (2.1 cm3 g(-1)). The pore diameter was 67 angstroms, so it was large enough to accommodate protein molecules inside the channels. Immobilization by physical adsorption of a commercial lipase preparation from Mucor javanicus was performed at different pH values (pH 5-8). pH 6 gave the highest lipase loading … Show more

“…[36] Inhibiting lipase activity by increasing the pH values is probably due to an unfavorable charge distribution on the amino acid residues, that produces a further activity decrease. [24] Since the highest hydrolytic activity was observed when lipase adsorption was conducted at pH 6.8, all other steps of this work were carried out under these conditions.…”

“…[17,22,23] These parameters have large influence on the total amount of enzyme loading and enzyme-catalyst activity. [24,25] Hydrophobic binding of lipases by adsorption has proved successful due to its affinity for water/oil interfaces. Thus, the present work deals with the synthesis of PS nanoparticles and their use as a support for the immobilization of Cal-B.…”

“…MCM-41 and SBA-15), possessing a hexagonal array of ordered mesopores has been widely investigated, partly due to the availability of large scale well-established synthetic protocols. [3,4] The corresponding cubic mesoporous silica having a cubic array of mesopores has the potential to be more efficient in catalysis and sorption processes, as the cubic array of pores is formed by a 3D cage-like system of interconnected pores providing easy accessibility in all directions. [5] However, cubic mesoporous silica has not been studied widely, as very controlled synthetic protocols are still lacking.…”
