Phylogenetics of Impatiens and Hydrocera (Balsaminaceae) Using Chloroplast atpB-rbcL Spacer Sequences

Abstract: ABSTRACT. Balsaminaceae are a morphologically diverse family with ca. 1,000 representatives that are mainly distributed in the Old World tropics and subtropics. To understand the relationships of its members, we obtained chloroplast atpB-rbcL sequences from 86 species of Balsaminaceae and five outgroups. Phylogenetic reconstructions using parsimony and Bayesian approaches provide a well-resolved phylogeny in which the sister group relationship between Impatiens and Hydrocera is confirmed. The overall topology … Show more

“…The close phylogenetic relationship of I. shimianensis to the four species mentioned above is The results of our phylogenetic analyses based on the ITS and atpB-rbcL information ( Figs. 3 , 4 ) were largely congruent with those of previous studies ( Yuan et al 2004 ;Janssens et al 2006 ;Yu 2008 ). In addition to the four species above, Yu (2008) also resolved I. chiulungensis Y. L. Chen to be a member of the clade referred to as Group 7 by Yu (2008) .…”

“…The sequences contained in the matrix accessions M2451.NX and M2452.NX ( Janssens et al 2006 ) in TreeBASE constituted the majority of the sequences used in this study. An additional 34 ITS and atpBrbcL sequences were downloaded from GenBank.…”

“…DNA was extracted for 62 species (Table 1) using a modified version of the hot CTAB protocol Janssens et al 2006). Specific primers used for the amplification of the C-terminal ends of the two AP3/DEF homologues in Impatiens (ImpDEF1 and ImpDEF2) were designed using the cloned cDNA sequences of Geuten et al (2006): IhCT-DEF1F (5 0 -GCAGATACACAGAAA CCTACTTCAG-3 0 ), IhCT-DEF1R (5 0 -TTATAGTTAAA GCAAAGCATAGGTTGTGAG-3 0 ), IhCT-DEF2F (5 0 -AC AGACACATAGAAACTTACTCCAGC-3 0 ), and IhCT-DEF2R (5 0 -CCATCACCATCTTCATCTTCAATCC-3 0 ).…”

“…According to Janssens et al (2006), I. omeiana is sister to all other Impatiens species and was, therefore, used as outgroup for all phylogenetic analyses. All sequences were submitted to GenBank (Table 1).…”

“…Subsequently, the exact phylogenetic relationship of the new species was determined by reanalyzing the clade to which I. nzabiana belongs. Primers and temperature profiles used for the amplification of atpB-rbcL , ImpDEF1 / ImpDEF2 and ITS follow Janssens et al (2006Janssens et al ( , 2007 and White et al (1990) , respectively. DNA extraction, PCR amplification, sequencing, sequence assembly and alignment were followed as described by Janssens et al (2008) .…”
