Molecular taxonomy often uncovers cryptic species, reminding us that taxonomic incompleteness is even more severe than previous thought. The importance of cryptic species for conservation is poorly understood. Although some cryptic species may be seriously threatened or otherwise important, they are rarely included in conservation programs as most of them remain undescribed. We analysed the importance of cryptic species in conservation by scrutinizing the South European cryptic complex of the subterranean amphipod Niphargus stygius sensu lato. Using uni- and multilocus delineation methods we show that it consists of 15 parapatric and sympatric species, which we describe using molecular diagnoses. The new species are not mere “taxonomic inflation” as they originate from several distinct branches within the genus and coexist with no evidence of lineage sharing. They are as evolutionarily distinct as average nominal species of the same genus. Ignoring these cryptic species will underestimate the number of subterranean endemics in Slovenia by 12 and in Croatia by four species, although alpha diversity of single caves remains unchanged. The new taxonomy renders national Red Lists largely obsolete, as they list mostly large-ranged species but omit critically endangered single-site endemics. Formal naming of cryptic species is critical for them to be included in conservation policies and faunal listings.

In this paper we supplement Greece's recent annotated inventory of freshwater fishes per hydrographic basin with recent distributional data and taxa alteration information, based on field sampling and a literature review up to September 2011. We report on newly documented distributional records of 31 fish species plus one unidentified taxon, within 35 hydrographic river basin units in Greece. These new records include 14 native fish species, seven alien and 12 translocated. Translocated taxa are distinguished from aliens, in order to report species non-indigenous to a basin but native within the same ecoregion. Twelve hydrographic basin units are newly added to the roster of ichthyologically explored river basins following a previous basin-scale inventory method (the total is now 117). This review increases the number of Greece's freshwater fish taxa to 167, since four new species are added to the list (Carassius langsdorfii, Neogobius fluviatilis, Telestes alfiensis, Millerigobius macrocephalus) and two are deleted (Salmo dentex, Barbus rebeli) due to taxonomic changes. Taxonomic changes will probably continue to alter the national list since phylogenetic research is ongoing on several taxa in many parts of the country.

In this research protein profiles of Popovo minnow muscle were presented and partially characterized for the first time. Delminichthys ghetaldii (Steindachner, 1882) is an endemic fish species which can be found in springs and streams in fields Ljubomir, Dabar and Fatnica in Bosnia and Herzegovina, and Bacina lakes in Croatia. Most of the year minnows dwell in underground water habitats of karst region in eastern Herzegovina. During the autumn and spring flooding of the surrounding fields, Popovo minnows emerge from the underground waters in estavele and spawn in newly formed temporary lakes. In order to obtain protein profiles from fish muscle, we used several extraction protocols and buffers. Sarcoplasmic proteins were extracted from muscle tissue using Tris/EDTA and Tris/CaCl2 buffers. The results showed that the most abundant sarcoplasmatic proteins had molecular weight of 40 and 34 kDa while high MW band appeared only in samples extracted by Tris/EDTA. Re-extraction with 1% SDS resolved the presence of myofibrillar proteins and showed to be qualitatively and quantitatively more efficient than the extraction by nonionic detergent Tween. Our results also indicate that protein extraction with commercial SDS buffer from a biopsy muscle tissue gives good screening results and is applicable for field work. The results are discussed in terms of the efficiency of different buffers in protein extraction from Popovo minnow muscle tissue.
